Three novel endo-β-N-acetylglucosaminidases (AVUL01, BCAC01, and BFIN01) classified as members of the glucoside hydrolase (GH) family 18 were identified from human fecal samples and then cloned and characterized for their ability to hydrolyze two ...

Nanomaterials offer dual applications in allergy management. For diagnosis, nanomaterials enhance analytical sensitivity and improve detection in specific IgE and functional assays such as the basophil activation test (BAT). For allergen‐specific immunotherapy, nanomaterials enable allergen masking and controlled release, and effectively modulate the ...

Immobilization of an Endo-β-N-acetylglucosaminidase for the Release of Bioactive N-glycans [PDF]
As more is learned about glycoproteins' roles in human health and disease, the biological functionalities of N-linked glycans are becoming more relevant. Protein deglycosylation allows for the selective release of N-glycans and facilitates glycoproteomic

A Cholesterol Analogue for Cell‐Surface Enzyme Display
We report a non‐genetic strategy for bacterial cell surface enzyme immobilisation using cholesterol‐based artificial lipids containing Ni2+‐NTA binding groups that can integrate into biological membranes. The engineered cells can capture His‐tagged enzymes directly from cell lysates while preserving intracellular and surface activity, allowing for ...

A fully synthetic four‐component vaccine has been developed combining Tn antigen, vizantin (Mincle agonist), rhamnose (antibody recruitment), and mannose (CD206 targeting). By synergistically activating Mincle, FcγR, and CD206 pathways, it elicits strong IgG switching and cytotoxic T‐cells, significantly inhibiting tumors in mice.

Tegument Glycoproteins and Cathepsins of Newly Excysted Juvenile Fasciola hepatica Carry Mannosidic and Paucimannosidic N-glycans. [PDF]
Recently, the prevalence of Fasciola hepatica in some areas has increased considerably and the availability of a vaccine to protect livestock from infection would represent a major advance in tools available for controlling this disease.

A reducible solubilizing tag strategy (RST‐2.0) enables the synthesis, purification and ligation of difficult peptides while being readily removable just during folding, which overcomes the common challenges from protein hydrophobicity and aggregation. With this method, we achieved the first total synthesis of native and N‐glycosylated IL‐15 as well as

Golgi self-correction generates bioequivalent glycans to preserve cellular homeostasis
Essential biological systems employ self-correcting mechanisms to maintain cellular homeostasis. Mammalian cell function is dynamically regulated by the interaction of cell surface galectins with branched N-glycans. Here we report that N-glycan branching

Porcine kidney xenotransplantation: From primate models to clinical reality
In the face of a critical shortage of human donor kidneys for end‐stage renal disease patients, porcine kidney xenotransplantation has emerged as a viable solution. This field has navigated major hurdles, including immune rejection, physiological incompatibilities, potential biomechanical differences and the risk of cross‐species infection. To overcome

We report plate‐based coupled assays to rapidly and quantitatively determine the activity and selectivity of mucin‐active carbohydrate sulfatases. This fluorogenic assay further enables inhibitor identification and allows determination of (sub‐)cellular location. ABSTRACT Sulfated glycans play a central role in human health and influence cell signaling,
