Studies on the network biology impact in pharmacology and toxicology

open access: yes, 2022
Network biology is a concept to deals with the biological process on a broader level. The molecular interactions at the cellular level are considered as a network. The aims of network biology are to investigate the components of cellular networks and their interactions. These approaches are widely used in pharmacology and toxicology.

Disrupting Helicobacter pylori Iron Homeostasis With Bismuth Nanodrug‐Mediated Nutritional Trap for Targeted Gastric Infection Therapy

open access: yesAdvanced Science, EarlyView.
A “nutritional trap” strategy centered on disrupting bacterial iron homeostasis is proposed. The synthesized Bi‐TP@FU (TBF) can target Helicobacter pylori, interfere with the bacterial iron uptake and utilization processes to disrupt its iron homeostasis, induce bacterial death, and meanwhile preserve the balance of the intestinal flora.

Molecular Dynamics‐Guided Sterol Engineering of mRNA‐Lipid Nanoparticles Reprograms Biodistribution and Enhances Spleen‐Selective Immunity

open access: yesAdvanced Science, EarlyView.
Sterol engineering enables precise control of mRNA lipid nanoparticles by tuning membrane organization. Replacing cholesterol with bile acid‐derived sterols redirects protein expression from the liver to the spleen. Notably, CA‐20 LNP enhances antigen‐specific immune responses while exhibiting a favorable safety profile, highlighting sterols as a ...

Analysis of Nicotine Toxicity and Mechanisms of Senescence in Nucleus Pulposus Cells Using Network Toxicology and Molecular Docking Technique

open access: yesJOR Spine
Aim Through the use of network toxicology, the research sought to determine whether cellular senescence and associated molecular mechanisms in nicotine‐induced intervertebral disc degeneration (IVDD) were potentially harmful. Methods The primary chemical

Identifying a Csmd3+ Microglial Subpopulation that Drives Cold‐to‐Hot Transition and Immune‐Cure in Glioblastoma

open access: yesAdvanced Science, EarlyView.
The study establishes an immune‐cure (ICu) mouse model based on a TMEMed G422TN‐GBM system that faithfully recapitulates human TMEMed GBM. scRNA‐seq analysis reveals a Csmd3+ microglial subset with innate immune memory (IIM) potential that potently suppresses GBM growth, drives a TME cold‐to‐hot transition, and induces 100% ICu in long‐term survival ...

Human Foetal Neuroblasts Exhibit BK Channel‐Dependent Membrane Voltage Oscillations upon Depolarization

open access: yesAdvanced Science, EarlyView.
Primary cultures of neuroblasts isolated from the nucleus basalis of Meynert of 12‐weeks‐old human foetuses were prepared. Whole‐cell patch‐clamp recordings were performed by injecting a depolarizing stimulus current (+500 pA; 500 ms), and the membrane voltage recorded; this stimulus current evoked periodic‐like oscillations in membrane voltage ...
