O-GlcNAc signaling increases neuron regeneration through one-carbon metabolism in Caenorhabditis elegans

open access: yeseLife
Cellular metabolism plays an essential role in the regrowth and regeneration of a neuron following physical injury. Yet, our knowledge of the specific metabolic pathways that are beneficial to neuron regeneration remains sparse. Previously, we have shown

Mitochondrial dysfunction remodels one-carbon metabolism in human cells

open access: yeseLife, 2016
Mitochondrial dysfunction is associated with a spectrum of human disorders, ranging from rare, inborn errors of metabolism to common, age-associated diseases such as neurodegeneration.

One-Carbon Metabolism: Pulling the Strings behind Aging and Neurodegeneration

open access: yesCells, 2022
One-carbon metabolism (OCM) is a network of biochemical reactions delivering one-carbon units to various biosynthetic pathways. The folate cycle and methionine cycle are the two key modules of this network that regulate purine and thymidine synthesis ...

S-Adenosylmethionine (SAM) and S-Adenosylhomocysteine (SAH) Monitoring Using Analytical Methods in Clinical Laboratory Practice: Where Are We?

open access: yesBiomedicines
S-adenosylmethionine (SAM) and S-adenosylhomocysteine (SAH) are essential intermediates in one-carbon metabolism and key regulators of cellular methylation capacity. Their concentrations and the SAM/SAH ratio are increasingly studied as biomarkers across

Metabolic responses of primary and transformed cells to intracellular Listeria monocytogenes. [PDF]

open access: yes, 2012
The metabolic response of host cells, in particular of primary mammalian cells, to bacterial infections is poorly understood. Here, we compare the carbon metabolism of primary mouse macrophages and of established J774A.1 cells upon Listeria monocytogenes

Gut microbiome and aging—A dynamic interplay of microbes, metabolites, and the immune system

open access: yesFEBS Letters, EarlyView.
Age‐dependent shifts in microbial communities engender shifts in microbial metabolite profiles. These in turn drive shifts in barrier surface permeability of the gut and brain and induce immune activation. When paired with preexisting age‐related chronic inflammation this increases the risk of neuroinflammation and neurodegenerative diseases.

Abnormalities in one-carbon metabolism in young patients with psychosis

open access: yesFrontiers in Psychiatry, 2023
IntroductionFolates, the main actors in one-carbon (C1) metabolism, are involved in synthesising monoamines and maintaining genomic stability. Previous studies support the association between C1 metabolism and schizophrenia.

Glycine decarboxylase deficiency causes neural tube defects and features of non-ketotic hyperglycinemia in mice. [PDF]

open access: yes, 2015
Glycine decarboxylase (GLDC) acts in the glycine cleavage system to decarboxylate glycine and transfer a one-carbon unit into folate one-carbon metabolism. GLDC mutations cause a rare recessive disease non-ketotic hyperglycinemia (NKH).

Reconstructing enzyme evolution by protein engineering

open access: yesFEBS Letters, EarlyView.
Natural enzyme evolution can be retraced by protein engineering methods such as directed evolution, rational design, and ancestral sequence reconstruction. These approaches reveal how enzymes emerged from ligand‐binding scaffolds, developed varying substrate preferences, formed oligomeric complexes, adapted to environmental changes, and evolved novel ...

Chromosomal Instability Causes Sensitivity to Polyamines and One-Carbon Metabolism

open access: yesMetabolites, 2023
Aneuploidy, or having a disrupted genome, is an aberration commonly found in tumours but rare in normal tissues. It gives rise to proteotoxic stress as well as a stereotypical oxidative shift, which makes these cells sensitive to internal and ...
