The translational research framework integrating TCM for RA. The figure outlines a stepwise framework for the modernization and clinical translation of TCM in RA. Part 1 highlights the inheritance and development of TCM theory, including classical medical texts, traditional herbal knowledge, and syndrome differentiation–based treatment principles. Part

Immunomodulatory Nanozymes as Programmable Redox–Immune Set‐Point Regulators
Conceptual phase space linking reactive oxygen species level/dynamics to immune state, with pathological set‐points for chronic inflammation, tumor escape, and degeneration contrasted against a central healthy redox–immune set‐point. Buffering and amplifying nanozymes are depicted as devices that dial redox tone up or down to steer diseased ...

This study elucidates a novel Kupffer cell‐neutrophil‐hepatocyte axis that drives metabolic‐immune crosstalk during acute liver failure. Specifically, lactate‐primed NETs release links metabolic reprogramming to NETosis‐induced regenerative failure in hepatocytes via the TLR9/KLF15/AJUBA axis.

Functional roles of the non-catalytic calcium-binding sites in the N-terminal domain of human peptidylarginine deiminase 4. [PDF]
This study investigated the functional roles of the N-terminal Ca(2+) ion-binding sites, in terms of enzyme catalysis and stability, of peptidylarginine deiminase 4 (PAD4).

Activated CCG10‐NLR: Octameric resistosome driving sustained calcium influx
This commentary spotlights landmark work from Guo et al. resolving the structure of the octameric resistosome formed by the active CCG10‐nucleotide‐binding leucine‐rich repeat (NLR) immune receptor WAI3, which triggers sustained, multi‐phasic Ca2+ influx.

Peptidomimetics as protein arginine deiminase 4 (PAD4) inhibitors
The protein arginine deiminase 4 (PAD4) is a calcium-dependent enzyme, which catalyses the irreversible conversion of peptidyl-arginines into peptidyl-citrullines and plays an important role in several diseases such as in the rheumatoid arthritis, multiple sclerosis, Alzheimer's disease, Creutzfeldt-Jacob's disease and cancer.

Probing the Roles of Calcium-Binding Sites during the Folding of Human Peptidylarginine Deiminase 4
Our recent studies of peptidylarginine deiminase 4 (PAD4) demonstrate that its non-catalytic Ca2+-binding sites play a crucial role in the assembly of the correct geometry of the enzyme. Here, we examined the folding mechanism of PAD4 and the role of Ca2+

The Arabidopsis E3 ligase SNIPER8 negatively regulates immunity by promoting ubiquitination and proteasomal degradation of the transcriptional corepressor TOPLESS‐RELATED1, limiting excessive immune responses, maintaining immune homeostasis, and balancing defense activation and normal plant growth.

Autoimmunity and Periodontitis
In a microbe‐driven inflammatory environment, peptidyl‐arginine deiminase (PAD) enzymes from neutrophils and Porphyromonas gingivalis citrullinate both microbial and self‐antigens. B cell presentation of citrullinated or self‐mimicking epitopes activates T cells that assist B cells in antibody isotype switching, affinity maturation, epitope spreading ...
