It is well established that the cyclic GMP‐dependent protein kinase I (PKG1) is canonically activated by cyclic guanosine monophosphate (cGMP), enabling its regulation of vascular tone, cardiac function and smooth muscle homeostasis. Gasdermin‐Mediated Pyroptosis: Novel Strategies Against Colorectal Cancer

Pyroptosis is a double‐edged sword in colorectal cancer (CRC): chronic low‐grade pyroptosis promotes tumor progression, whereas acute induction of pyroptosis in cancer cells triggers antitumor immunity and enhances therapeutic response. Targeting gasdermin‐mediated pyroptosis represents a promising strategy for CRC treatment.

Organ‐Specific Cancer‐Associated Fibroblast Subtypes Across the Digestive System Linked to Cancer Hallmarks

We integrated 14 single‐cell RNA‐seq datasets and 18 bulk cohorts to define eight CAF subtypes in GI cancers. myCAF1 and myCAF2 promoted immunosuppressive, poor‐prognosis niches, whereas iCAF1 was linked to favorable prognosis and antitumor immunity.
