In Vitro Selection of Functional Neoproteins Containing Highly Fluorinated Cores

open access: yesAngewandte Chemie, EarlyView.
The generation of neoproteins—functional proteins containing unnatural amino acids—is a key goal in synthetic biology. We develop a platform to discover neoproteins with specified bioactivity from trillion‐member sequence libraries, using genetic code reprogramming, in vitro selection and microwave peptide synthesis.

A Dual‐Delivery System Designed to Overcome EV‐Mediated Ferroptosis Resistance for Efficient Solid Tumor Therapy

open access: yesAdvanced Science, EarlyView.
(A) Establishment and characterization of the osteosarcoma‐targeted dual‐delivery nanoplatform mBAP1/siENO1@LNP‐OTP for simultaneous regulation of BAP1 and ENO1 in tumor cells. (B) After intravenous administration, mBAP1/siENO1@LNP‐OTP preferentially accumulates at tumor sites through OTP‐mediated targeting.

Designing phage cocktails to combat the emergence of bacteriophage-resistant mutants in multidrug-resistant Klebsiella pneumoniae

open access: yesMicrobiology Spectrum
Phage therapy, an emerging strategy in the fight against recalcitrant bacterial infections, encounters a significant hurdle posed by the development of phage-resistant bacterial strains. To address this concern, we suggest a novel phage cocktail.

A Chiral Virus‐Like Nanoplatform That Broadly Mediates Antibiotic Delivery to the Brain for Bacterial Meningitis Therapy

open access: yesAdvanced Science, EarlyView.
Chiral virus‐mimicking mesoporous silica nanostructures (CVSNs) efficiently transport diverse antibiotics across the blood–brain barrier (BBB) into brain parenchyma and cerebrospinal fluid. By enhancing delivery to microglia and astrocytes, CVSNs improve antibiotic bioavailability and therapeutic efficacy against both Gram‐negative and Gram‐positive ...

Quantifying Structure‐Dependent Rigidity of 3D DNA Origami Boxes

open access: yesAdvanced Intelligent Systems, EarlyView.
We investigate the mechanical stability of a hollow 3D DNA origami box during assembly using liquid AFM imaging and finite element simulations. As structural completeness increases from flat sheets through intermediate assemblies to a closed box, rigidity improves.

Selection of porin and LPS mutants during infection by phage T4 facilitates the emergence of β-lactam resistance in Escherichia coli

open access: yesnpj Antimicrobials and Resistance
Understanding all sources of selective pressure that contribute to the emergence of antibiotic resistance is essential for developing sustainable antimicrobial strategies. Here, we investigated the interaction between phage T4 and Escherichia coli MG1655

Complete Pathway Refactoring and Combinatorial Biosynthesis of Anthracyclines for Modulation of Histone Eviction and DNA Damage Bioactivities

open access: yesAngewandte Chemie, EarlyView.
A synthetic biology platform was established for the complete refactoring of complex anthracycline biosynthetic pathways in Streptomyces coelicolor using standardized, chemically synthetic DNA parts. This modular construction enabled streamlined combinatorial biosynthesis, yielding novel analogs that decouple DNA damage from chromatin damage (histone ...

Using phage to drive selections toward restoring antibiotic sensitivity in Pseudomonas aeruginosa via chromosomal deletions

open access: yesFrontiers in Microbiology
Phage therapy has re-emerged in modern medicine as a robust antimicrobial strategy in response to the increasing prevalence of antimicrobial-resistant bacteria. However, bacterial resistance to phages can also arise via a variety of molecular mechanisms.
