Recent Insights in Multi‐Target Drugs in Pharmacology and Medicinal Chemistry

open access: yesChemMedChem, EarlyView.
This review highlights the rationale behind multitarget drug design as a promising approach to address diseases with complex etiologies. By combining pharmacophore features from different single‐target drugs, multitarget compounds can interact with multiple biological targets simultaneously.

Development of Keap1‐Nrf2 Protein–Protein Interaction Inhibitor Activating Intracellular Nrf2 Based on the Naphthalene‐2‐acetamide Scaffold, and its Anti‐Inflammatory Effects

open access: yesChemMedChem, EarlyView.
Pyrrolidine‐type naphthalene‐2‐acetamide compound 5i inhibits Kelch‐like ECH‐associated protein 1 (Keap1)‐nuclear factor erythroid 2‐related factor 2 (Nrf2) protein–protein interaction by binding to the Keap1‐DC domain; it subsequently activates intracellular Nrf2 and induces Nrf2‐target genes.

Synthesis of Urea‐Containing Derivatives and their Application as Potential Anti‐Methicillin‐Resistant Staphylococcus Aureus Agents

open access: yesChemMedChem, EarlyView.
A series of propylureido amides against methicillin‐resistant Staphylococcus aureus (MRSA) activity was prepared. The design involved replacing other hydrogen bond donors reported in FabI inhibitors with a urea moiety, while maintaining other pharmacophoric features.
