Gasdermin‐Mediated Pyroptosis: Novel Strategies Against Colorectal Cancer
Pyroptosis is a double‐edged sword in colorectal cancer (CRC): chronic low‐grade pyroptosis promotes tumor progression, whereas acute induction of pyroptosis in cancer cells triggers antitumor immunity and enhances therapeutic response. Targeting gasdermin‐mediated pyroptosis represents a promising strategy for CRC treatment.

Abstract Background The combination of olaparib plus radium‐223 improved progression‐free survival in patients with metastatic castration‐resistant prostate cancer with bone metastases (BM) in the multicenter, randomized, phase 2 COMRADE trial (NCT03317391). Here, the patient‐reported outcome (PRO) analysis of this trial is reported.

What's New? Epithelial ovarian cancer (EOC) is a severe malignancy with few effective treatment options. While the mechanisms remain unclear, changes in lipid metabolism may fuel tumor growth and EOC progression, offering a possible path for new treatments.

Controversies and clinical unknowns in the use of PARP inhibitors in ovarian cancer
Poly (adenosine diphosphate-ribose) polymerase inhibitors (PARPi) have significantly improved the treatment of advanced ovarian cancer, however, there are still many aspects of their use that require further understanding.

ABSTRACT Introduction Patients with ovarian cancer often present with massive ascites, leading to significant protein loss during surgical procedures. Although cell‐free concentrated ascites reinfusion therapy (CART) is used in palliative settings to mitigate protein loss, its application in intraoperative settings remains unexplored.

First-line treatment of advanced ovarian cancer: an expert update
Ovarian cancer is the fifth most common cause of death among malignant diseases in women in Europe. The standard treatment is cytoreductive surgery, followed by platinum-taxane based chemotherapy.

Bovine thymus poly(adenosine diphosphate ribose) polymerase.
About 1,300-fold purification of poly(adenosine diphosphate ribose) polymerase has been achieved from the extract of bovine thymus with a recovery of 10 to 20%. The final preparation has a purity of 99%, and the enzyme is composed of a single peptide with a molecular weight of 130,000. The purified enzyme required NAD+, Mg2+, a thiol compound, DNA, and

Plecstatin inhibits hepatocellular carcinoma tumorigenesis and invasion through cytolinker plectin
The ruthenium‐based metallodrug plecstatin exerts its anticancer effect in hepatocellular carcinoma (HCC) primarily through selective targeting of plectin. By disrupting plectin‐mediated cytoskeletal organization, plecstatin inhibits anchorage‐dependent growth, cell polarization, and tumor cell dissemination.

Abstract Nicotinamide adenine dinucleotide (NAD+) is an essential metabolite involved in numerous biological processes, functioning both as a redox cofactor in cellular energy metabolism and as a co‐substrate for NAD+‐utilizing enzymes, including sirtuins and poly(ADP‐ribose) polymerases.
