Polymyxin B-Induced Kidney Injury Assessment of a Novel Formulation of Polymyxin B (VRP-034) in Rats [PDF]

open access: yes, 2021
Despite the crucial role of Polymyxin-B in treating life-threatening gram-negative infections, its clinical utility is limited due to the risk of acute kidney injury.

Predictors of polymyxin B treatment failure in Gram-negative healthcare-associated infections among critically ill patients

open access: yesJournal of Microbiology, Immunology and Infection, 2018
Background: With increasing prevalence and spread of multidrug resistant Gram-negative infections, parenteral polymyxins resurged in clinical practice.

Polymyxin Induces Significant Transcriptomic Perturbations of Cellular Signalling Networks in Human Lung Epithelial Cells

open access: yesAntibiotics, 2022
Inhaled polymyxins are increasingly used to treat pulmonary infections caused by multidrug-resistant Gram-negative pathogens. We have previously shown that apoptotic pathways, autophagy and oxidative stress are involved in polymyxin-induced toxicity in ...

Inactivation of Endotoxin by Polymyxin B [PDF]

open access: yesAntimicrobial Agents and Chemotherapy, 1974
The limulus gelation assay was utilized to investigate endotoxin inactivation by a number of antibiotics in vitro. Endotoxin activity was sharply reduced by polymyxin B and sodium colistimethate. The effect of the polymyxin was not significantly inhibited by 0.001 M calcium or 90% serum.

Personalizing Polymyxin B dosing using an adaptive feedback control algorithm [PDF]

open access: yes, 2018
Polymyxin B is used as an antibiotic of last resort for patients with multidrug-resistant Gram-negative bacterial infections; however, it carries a significant risk of nephrotoxicity.

POLYMYXIN B AND HEART MUSCLE

open access: yesBritish Journal of Anaesthesia, 1979
The effects of the polypeptide antibiotic, polymyxin B, on myocardial contractility were studied in t;e isolated rat heart muscle. Five different doses of polymyxin B were tested. There were no changes in contractility with does ranging from the clinical therapeutic value to three times greater.

Novel polymyxin derivatives are less cytotoxic than polymyxin B to renal proximal tubular cells [PDF]

open access: yes, 2012
The emergence of very multiresistant Gram-negative bacterial strains has reinstated polymyxins (polymyxin B, colistin), pentacationic lipopeptides, in the therapy, in spite of their nephrotoxicity. Extensive tubular reabsorption concentrates polymyxin in

Emergence of polymyxin B-heteroresistant hypervirulent Klebsiella pneumoniae from an individual in the community with asymptomatic bacteriuria

open access: yesBMC Microbiology, 2022
Background The heteroresistance of polymyxin B, a last-resort antibiotic used to treat many serious bacterial infections, may lead to antibiotic treatment failure.
