Orally administered melatonin prevents lipopolysaccharide-induced neural tube defects in mice. [PDF]

open access: yesPLoS ONE, 2014
Lipopolysaccharide (LPS) has been associated with adverse pregnant outcomes, including fetal demise, intra-uterine growth restriction (IUGR), neural tube defects (NTDs) and preterm delivery in rodent animals.

The promise and challenges of exploiting the proton-coupled folate transporter for selective therapeutic targeting of cancer. [PDF]

open access: yesCancer Chemother Pharmacol, 2018
This review considers the "promise" of exploiting the proton-coupled folate transporter (PCFT) for selective therapeutic targeting of cancer. PCFT was discovered in 2006 and was identified as the principal folate transporter involved in the intestinal absorption of dietary folates.

Structure Function Studies of the Proton Coupled Folate Transporter [PDF]

open access: yesBiophysical Journal, 2010
Folate vitamins are essential for DNA replication and cellular proliferation. However, mammalian organisms are devoid of folate biosynthesis and rely on dietary sources to meet their metabolic requirement for folate cofactors. The proton coupled folate transporter (PCFT/SLC46A1) has been recently identified as the molecular entity responsible for ...

Novel localization of folate transport systems in the murine central nervous system

open access: yesFluids and Barriers of the CNS, 2022
Background Folates are a family of B9 vitamins that serve as one-carbon donors critical to biosynthetic processes required for the development and function of the central nervous system (CNS) in mammals.

Protective effects of pyrroloquinoline quinone in brain folate deficiency

open access: yesFluids and Barriers of the CNS, 2023
Background Folates (Vitamin B9) are critical for normal neurodevelopment and function, with transport mediated by three major pathways: folate receptor alpha (FRα), proton-coupled folate transporter (PCFT), and reduced folate carrier (RFC).

Stereoselective Transport of Amethopterin Enantiomers by the Proton-coupled Folate Transporter

open access: yesDrug Metabolism and Pharmacokinetics, 2010
Stereoselective transport of methotrexate (L-amethopterin, L-MTX) and its antipode (D-amethopterin, D-MTX) by the proton-coupled folate transporter (PCFT) was examined using PCFT-expressing HEK293 cells (PCFT-HEK293 cells). Uptake of both L-MTX and D-MTX was pH-dependent and decreased with an increase in the extracellular pH from 5.0 to 7.4.

Luteolin Enhances Choroid Plexus 5-MTHF Brain Transport to Promote Hippocampal Neurogenesis in LOD Rats

open access: yesFrontiers in Pharmacology, 2022
Folates, provided by food, are commonly used antidepressant synergists in late-onset depression (LOD). However, increased intake of folic acid in the elderly population might lead to the accumulation of unmetabolized folic acid in the systemic ...

Novel insights in folate receptors and transporters: implications for disease and treatment of immune diseases and cancer

open access: yesPteridines, 2015
Folate receptors and transporters as well as folate enzymes play an essential role in human disease and form important targets for the treatment of immune diseases and cancer.

Targeted therapy of pyrrolo[2,3-d]pyrimidine antifolates in a syngeneic mouse model of high grade serous ovarian cancer and the impact on the tumor microenvironment

open access: yesScientific Reports, 2022
Novel therapies are urgently needed for epithelial ovarian cancer (EOC), the most lethal gynecologic malignancy. In addition, therapies that target unique vulnerabilities in the tumor microenvironment (TME) of EOC have largely been unrealized.
