Embryo‐Derived Cathepsin B Promotes Implantation and Decidualization by Activating Pyroptosis

open access: yesAdvanced Science
Embryo implantation and decidualization are crucial for a successful pregnancy. How the inflammatory response is regulated during these processes is undefined. Pyroptosis is an inflammatory form of cell death mediated by gasdermin D (GSDMD).

The role of pyroptosis-related genes in sepsis.

open access: yes, 2023
(A)The Sankey plot reveals relationship between pyroptosis-related clusters, pyroptosis-related gene clusters, and pyroptosis-related score groups. (B-C) The pyroptosis-related score in different pyroptosis-related clusters and pyroptosis-related gene ...

Brown Adipocyte Sheets Alleviate Myocardial Ischemia–Reperfusion Injury Through NRG4–ErbB4–Dependent Ferroptosis Inhibition

open access: yesAdvanced Science, EarlyView.
Brown adipocyte sheets are engineered to protect the heart against myocardial ischemia–reperfusion injury by restraining ferroptosis. Upon transplantation onto the cardiac surface, they improve cardiac function, limit infarction and fibrosis, and enhance angiogenesis.

PIEZO1 is Required for Acute Myeloid Leukemia Progression and Leukemia Stem Cell Maintenance via HIF1A‐SLC7A11 Axis‐Mediated Ferroptosis Defense

open access: yesAdvanced Science, EarlyView.
This study identifies the mechanosensor PIEZO1 as a key factor in promoting acute myeloid leukemia progression and uncovers a ferroptosis defense system mediated by PIEZO1‐HIF1A‐SLC7A11 axis that is essential for leukemia stem cell maintenance. This ferroptotic defense system may represent a unique vulnerability for leukemia stem cells, providing a ...

Pyroptosis in Cancer: Friend or Foe?

open access: yes, 2021
Pyroptosis is an inflammatory form of programmed cell death that is mediated by pore-forming proteins such as the gasdermin family (GSDMs), including GSDMA-E. Upon cleavage by activated caspases or granzyme proteases, the N-terminal of GSDMs oligomerizes

PANX2 Suppresses Lung Adenocarcinoma Progression by Inducing Disulfidptosis and Enhancing Antitumor Immunity

open access: yesAdvanced Science, EarlyView.
ABSTRACT Lung adenocarcinoma (LUAD) remains a leading cause of cancer mortality with limited therapeutic options. Disulfidptosis, a novel cell death modality driven by disulfide stress, represents a promising target, yet its regulation in LUAD is poorly defined. Here, we identify Pannexin 2 (PANX2) as a tumor suppressor in LUAD.

Induction of Pyroptosis and Its Implications in Cancer Management

open access: yesFrontiers in Oncology, 2019
Pyroptosis is a gasdermins mediated programmed cell death, which has been widely studied in inflammatory disease models. Recently, there are growing evidences that pyroptosis can be chemically induced in cancer cells without any bacterial or viral ...

ASC filament formation serves as a signal amplification mechanism for inflammasomes [PDF]

open access: yes, 2016
A hallmark of inflammasome activation is the ASC speck, a micrometre-sized structure formed by the inflammasome adaptor protein ASC (apoptosis-associated speck-like protein containing a CARD), which consists of a pyrin domain (PYD) and a caspase ...

Triggering Pyroptosis in Cancer

open access: yesBiomolecules
Pyroptosis is an inflammatory programmed cell death recently identified as a crucial cellular process in various diseases, including cancers. Unlike other forms of cell death, canonical pyroptosis involves the specific cleavage of gasdermin by caspase-1, resulting in cell membrane damage and the release of the pro-inflammatory cytokines IL-1β and IL-18.
