Resolving the homology-function relationship through comparative genomics of membrane-trafficking machinery and parasite cell biology [PDF]
With advances in DNA sequencing technology, it is increasingly common and tractable to informatically look for genes of interest in the genomic databases of parasitic organisms and infer cellular states.

RAB7 deficiency impairs pulmonary artery endothelial function and promotes pulmonary hypertension
Pulmonary arterial hypertension (PAH) is a devastating and progressive disease with limited treatment options. Endothelial dysfunction plays a central role in the development and progression of PAH, yet the underlying mechanisms are incompletely ...

Modulation of Rab5 and Rab7 recruitment to phagosomes by phosphatidylinositol 3-kinase [PDF]
Phagosomal biogenesis is central for microbial killing and antigen presentation by leukocytes. However, the molecular mechanisms governing phagosome maturation are poorly understood.

Skeletal muscle fibrosis stems from abnormal extracellular matrix accumulation due to excessive production or reduced degradation of collagen. Obesity induces this fibrosis, along with muscle atrophy and impaired regeneration. Nur77, a transcription factor encoded by NR4A1, is involved in diverse life activities.

A new Rab7 effector controls phosphoinositide conversion in endosome maturation [PDF]
Endosome maturation requires a coordinated change in the Rab GTPase and phosphoinositide composition of the endosomal membrane. In this issue, Liu et al. (2017. J. Cell Biol. https://doi.org/10.1083/jcb.201705151) identify WDR91 as a ubiquitous Rab7 effector that inhibits phosphatidylinositol 3-kinase activity on endosomes and is critical for endosome ...

Specific recycling receptors are targeted to the immune synapse by the intraflagellar transport system [PDF]
T cell activation requires sustained signaling at the immune synapse (IS), a specialized interface with the APC that assembles following TCR engagement by MHC-bound peptide.

CCDC41 is essential for meiotic maturation in mouse oocytes through regulating Rab7‐positive endosomes fusion with lysosomes and Rab11a‐positive vesicle fusion with the plasma membrane. Abstract Coiled‐coil domain‐containing protein 41 (CCDC41), a core component of centriolar distal appendages involved in centriole assembly and ciliary vesicle docking,

Class III PI3K regulates organismal glucose homeostasis by providing negative feedback on hepatic insulin signalling. [PDF]
Defective hepatic insulin receptor (IR) signalling is a pathogenic manifestation of metabolic disorders including obesity and diabetes. The endo/lysosomal trafficking system may coordinate insulin action and nutrient homeostasis by endocytosis of IR and ...

This study introduces a biomimetic nanoplatform (MTCA@C) designed for precise cancer therapy. It targets mitochondria to induce dysfunction through photodynamic activation, while concurrently blocking mitophagy via lysosomal alkalinization. The platform further triggers methuosis, resulting in tumor cell death through vacuolization and energy pathway ...

Human mesenchymal stem cells conditionally secrete sonic hedgehog (SHH)‐containing extra‐large extracellular vesicles (XLEVs) upon PI3K activation, which enriches the GDP‐bound form of Rab18. Rab18‐GDP drives perinuclear maturation and polarized vertical release of SHH‐XLEVs via Hsp90α‐ and nSMase2‐dependent mechanisms.
