In the course of suppression subtractive hybridization between the cDNA of phytohemagglutinin-stimulated and non-stimulated head kidney cells in rainbow trout (Oncorhynchus mykiss), a cDNA clone was obtained that showed most similarity to mammalian receptor for the anaphylatoxin of the fifth complement component (C5aR).

Expression of Receptors for Anaphylatoxins C3a and C5a on Rat Islet Preparations
Transplantation Proceedings, 2011Complement activation has been implicated in the development of the instant blood-mediated inflammatory reaction (IBMIR). In particular, anaphylatoxins C3a and C5a elicit a broad range of proinflammatory effects, including chemotaxis of inflammatory cells and cytokine release.

Identification and synthesis of a receptor binding site of human anaphylatoxin C5a
Journal of Medicinal Chemistry, 1991C5a is a 74 amino acid polypeptide that likely plays an important role in the pathogenesis of a number of inflammatory diseases. Therefore, the discovery of a C5a antagonist is of considerable therapeutic interest. A series of peptides designed to survey various regions of the molecule was synthesized by solid-phase peptide synthesis and evaluated for ...

e21084 Background: Accumulated evidence has revealed implications of chemokines and their receptors in the pathophysiology of cancer. Anaphylatoxin C5a, a fragment of the complement 5th component (C5), is a potent chemokine for leukocytes that express the C5a receptor (C5aR).

Cellular receptors to the anaphylatoxins C3a and C5a.
Biochemical Society symposium, 1987A single component in the plasma membrane of human polymorphonuclear leukocytes (hPMN) has been identified as the binding site for C5a. Labelled human C5a can be cross-linked to a 48,000-Mr membrane component on the hPMN surface by using the bifunctional reagent ethylene glycol bis(succinimidyl succinate).

Anaphylatoxin C5a modulation of an alpha-adrenergic receptor system in the rat hypothalamus
Journal of Neuroimmunology, 1985C5a anaphylatoxin injected via implanted cannulae into the perifornical region of the hypothalamus stimulated eating in sated rats. C5a also attenuated carbamyl choline-induced drinking, and carbamyl choline inhibited C5a-induced eating, a mutual inhibition characteristic of the adrenergic-cholinergic interactions at this site.

Expression cloning of a receptor for C5a anaphylatoxin on differentiated HL-60 cells
Biochemistry, 1991A cDNA clone encoding the human C5a anaphylatoxin receptor has been isolated by expression cloning from a CDM8 expression library prepared from mRNA of human myeloid HL-60 cells differentiated to the granulocyte phenotype with dibutyryladenosine cyclic monophosphate.

We have constructed a novel recombinant C5a anaphylatoxin (C5a-N19) containing a 19-residue amino-terminal extension peptide, using a plasmid vector which secretes the nascent polypeptide to the Escherichia coli periplasmic space. C5a-N19 was purified from cell lysates by immunoaffinity chromatography using a monoclonal antibody which recognizes a ...

Expression of the complement C5a anaphylatoxin receptor (C5aR) on non-myeloid cells
Immunology Letters, 1995The expression of the complement C5a anaphylatoxin receptor (C5aR) has traditionally been thought to be limited to myeloid blood cells, including neutrophils, monocytes, macrophages, and eosinophils. Immunohistochemistry and ligand-binding studies reported here demonstrate, however, that C5aR is expressed by parenchymal cells of several solid organs ...

Plasma clearance of the human C5a anaphylatoxin by binding to leucocyte C5a receptors.
Immunology, 1995The C5a anaphylatoxin is a potent complement-derived mediator of inflammation with chemotactic activity. In this study the possible role of specific high-affinity binding sites for C5a on peripheral blood leucocytes for the removal of C5a from human blood plasma was investigated.
