TrkA Receptor Endolysosomal Degradation is Both Ubiquitin and Proteasome Dependent [PDF]

open access: yesTraffic, 2008
Gaps in our knowledge exist regarding the degradation of the tropomyosin‐regulated kinase A (TrkA) receptor after addition of neurotrophin, nerve growth factor (NGF). TrkA is rapidly and transiently ubiquitinated upon addition of NGF.

Constitutive TrkA Activity in Receptor-Overexpressing PC12 Clones

Biochemical and Biophysical Research Communications, 2002
We have studied ligand-independent signaling by the nerve growth factor receptor TrkA in PC12 clones, under conditions of receptor overexpression. Our results indicate that TrkA-overexpressing PC12 clones display constitutive receptor activation, involving both the mature, 140-kDa form and the immature, intracellular 110-kDa form of the receptor ...

Tracking TrkA’s Trafficking: NGF Receptor Trafficking Controls NGF Receptor Signaling

Molecular Neurobiology, 2007
Growth factors such as the neurotrophins promote neuronal survival and shape neuronal morphology. Neurotrophin receptors are located on the surface of axons and dendrites and must convey their signal retrogradely to the nucleus to influence transcription of target genes. The distance between the site of receptor activation and the nucleus is tremendous.

Rat visual cortical neurones express TrkA NGF receptor

Neuroreport, 2002
In this study we report the expression of TrkA receptor within the rat visual cortex during postnatal development and in adulthood, using a specific monoclonal antibody which recognizes the extracellular domain of TrkA receptor. TrkA was not detected by immunohistochemistry at postnatal day 13 (P13), i.e. before eye opening.

Selectively Targeting Tropomyosin Receptor Kinase A (TRKA) via PROTACs

Journal of Medicinal Chemistry, 2020
Tropomyosin receptor kinases (TRKs) are promising cancer therapeutic targets. Chen et al. (J. Med. Chem. 2020, DOI: 10.1021/acs.jmedchem.0c01342) report the discovery of CG416 and CG428 as two potent small-molecule proteolysis-targeting chimera (PROTAC) degraders selective for TRKA over TRKB and TRKC.

Expression of neurotrophin receptor TrkA inhibits angiogenesis in neuroblastoma

Medical and Pediatric Oncology, 2000
Mechanisms regulating the expression of angiogenic factors in tumor cells are largely unknown. High expression of the neurotrophin receptor TrkA in neuroblastomas (NB) is associated with favorable prognosis, whereas TrkB is expressed on aggressive, MYCN-amplified NB.To investigate the biological effects of TrkA and TrkB expression on angiogenesis in NB,

Tetraspanin1 promotes NGF signaling by controlling TrkA receptor proteostasis

Cellular and Molecular Life Sciences, 2019
The molecular mechanisms that control the biosynthetic trafficking, surface delivery, and degradation of TrkA receptor are essential for proper nerve growth factor (NGF) function, and remain poorly understood. Here, we identify Tetraspanin1 (Tspan1) as a critical regulator of TrkA signaling and neuronal differentiation induced by NGF.
