Action of Natural Products on P2 Receptors: A Reinvented Era for Drug Discovery

open access: yesMolecules, 2012
Natural products contribute significantly to available drug therapies and have been a rich source for scientific investigation. In general, due to their low cost and traditional use in some cultures, they are an object of growing interest as alternatives

Contributions of purinergic P2X3 receptors within the midbrain periaqueductal gray to diabetes-induced neuropathic pain

open access: yesThe Journal of Physiological Sciences, 2015
Hyperalgesia and allodynia are commonly observed in patients with diabetic neuropathy. The mechanisms responsible for neuropathic pain are not well understood. Thus, in this study, we examined the role played by purinergic P2X3 receptors of the midbrain periaqueductal gray (PAG) in modulating diabetes-induced neuropathic pain because this brain region ...

Nutrient‐specific combinatorial secretion of GLP‐1 and neurotransmitters in the enteroendocrine L‐cell line GLUTag

open access: yesThe FEBS Journal, EarlyView.
Enteroendocrine L cells function as chemosensors in the intestine. Using the L‐cell line GLUTag, we show that distinct nutrients trigger different combinations of co‐secretion of GLP‐1 and neurotransmitters: D‐glucose evokes GLP‐1, glutamate, and ATP; L‐glutamine evokes GLP‐1 and glutamate; sucralose and butyrate evoke only GLP‐1.

Topical ATP Application in the Peripheral Swallowing-Related Regions Facilitates Triggering of the Swallowing Reflex Involving P2X3 Receptors

open access: yesFunction
The swallowing reflex is a critical component of the digestive process, triggered when food or liquids pass from the oral cavity to the oesophagus. Although adenosine triphosphate (ATP) is involved in various physiological processes, its potential to ...

Synergistic Effects of Bacterial Mimetic and Mechanical Stimulation in Dental Pulp Cell Inflammatory Signalling: A Laboratory Investigation

open access: yesInternational Endodontic Journal, EarlyView.
ABSTRACT Objectives Mechanical and/or microbial stimuli can prompt extracellular ATP release in many cell types, where it acts as an inflammatory mediator to amplify pain signals and inflammation. This study aimed to investigate the synergistic effects of microbial and/or mechanical stimulation on ATP and cytokine release from dental pulp cells (DPCs ...

262 MODULATION OF BLADDER AFFERENT SIGNALS IN NORMAL AND SPINAL CORD INJURED RATS BY P2X3 AND P2X2/3 PURINERGIC RECEPTORS [PDF]

open access: yesJournal of Urology, 2012
plete spinal cord transection (spinal cord injured: SCI) at the T8/T9 level or a sham procedure (nonlesion: NL) were performed. Four weeks later, rats were again anesthetized with isofluorane, a suprapubic and a vein catheter were implanted and tracheotomy was performed for artificial ventilation. Laminectomy was executed at the L2 spinal level.

Anatomical and functional mapping of vagal nociceptive sensory nerve subsets innervating the mouse lower airways by intersectional genetics

open access: yesThe Journal of Physiology, EarlyView.
Abstract figure legend We used an intersectional approach with recombinase‐expressing mice and adeno‐associated virus to map and modulate distinct nociceptive afferents in the vagal ganglia. TRPV1+P2X2+ neurons resided in the nodose ganglion (N), innervated the lungs (many projected into the alveoli) but not the trachea, and projected to the nucleus ...

High‐altitude headache: Insights into pathophysiology and potential treatment implications

open access: yesThe Journal of Physiology, EarlyView.
Abstract figure legend High‐altitude headache occurs during prolonged exposure to low oxygen, such as during ascent to high altitude. Several risk factors increase its likelihood, with the degree of hypoxia, often driven by rapid ascent to greater altitudes, being the most prominent factor.

Activation of Adenosine Triphosphate-Gated Purinergic 2 Receptor Channels by Transient Receptor Potential Vanilloid Subtype 4 in Cough Hypersensitivity

open access: yesBiomolecules
Background: Transient receptor potential vanilloid subtype 4 (TRPV4) is a Ca2+-permeable non-selective cation channel that is involved in the development of cough hypersensitivity. Purinergic 2 receptors (P2X) belong to a class of adenosine triphosphate (

A taste for ATP: neurotransmission in taste buds

open access: yesFrontiers in Cellular Neuroscience, 2013
Not only is ATP a ubiquitous source of energy but it is also used widely as an intercellular signal. For example, keratinocytes release ATP in response to numerous external stimuli including pressure, heat and chemical insult. The released ATP activates
