PRDM1+ Malignant Cells Mediate an Immunosuppressive Landscape and Resistance to Neoadjuvant Chemoradiotherapy and Immunotherapy in Esophageal Squamous Cell Carcinoma

open access: yesAdvanced Science, EarlyView.
Integrated scRNA‐seq, scTCR‐seq analysis, and functional assays identify PRDM1+ malignant epithelial cells with hyper lipid peroxidation characteristics that demonstrate reduced responsiveness to the nICRT treatment. Principal factor PRDM1 activates cysteine metabolism genes to modulate lipid peroxidation (an intrinsic cellular pathway related to ...

Research Progress on the Regulation Mechanism of Senescence-associated Secretory Phenotype in Osteoporosis [PDF]

open access: yesZhongguo quanke yixue
Senescence-associated secretory phenotype (SASP) is an important feature of cellular senescence and plays an important role in regulating the disease microenvironment.

Atrophic Skeletal Muscle‐Derived Extracellular Vesicles Transfer miR‐125a‐5p to Inhibit Bone Formation in Osteoporosis during Aging

open access: yesAdvanced Science, EarlyView.
A muscle‐bone endocrine pathway in aging is revealed in which extracellular vesicles released from atrophic skeletal muscle (Aged‐SKM‐EVs) inhibit bone formation. These EVs deliver miR‐125a‐5p to osteoblasts, thereby suppressing the SIRT7‐Sp7 signaling axis and osteogenic differentiation.

Targeting Lactate and Lactylation in Cancer Metabolism and Immunotherapy

open access: yesAdvanced Science, EarlyView.
Lactate, once deemed a metabolic waste, emerges as a central regulator of cancer progression. This review elucidates how lactate and its epigenetic derivative, protein lactylation, orchestrate tumor metabolism, immune suppression, and therapeutic resistance.

NAD+ metabolism controls inflammation during senescence

open access: yesMolecular & Cellular Oncology, 2019
We have recently discovered that nicotinamide adenine dinucleotide metabolism controls the pro-inflammatory senescence-associated secretory phenotype during cellular senescence.

An Insulin‐Exosome‐TNFAIP8 Axis Drives Stromal Fibrosis and Therapeutic Resistance in Pancreatic Cancer

open access: yesAdvanced Science, EarlyView.
In pancreatic ductal adenocarcinoma (PDAC), insulin is associated with activation of PI3K/AKT–RAB3A signaling and enhanced secretion of TNFAIP8‐enriched exosomes from tumor cells. Uptake of these exosomes by fibroblasts is linked to TRIM21‐dependent STAT1 degradation and the emergence of myofibroblastic CAF–associated features, accompanied by increased

Activated Platelet–Released Heat Shock Protein 90α Triggers Autophagy‐Dependent Neutrophil Extracellular Trap Formation and Amplifies Sepsis

open access: yesAdvanced Science, EarlyView.
HSP90α is significantly upregulated in platelets from sepsis patients, with its origin from megakaryocyte‐derived trafficking. Furthermore, activated platelets secrete HSP90α into the extracellular space in both free and exosome‐associated forms. Finally, extracellular HSP90α directly engages TLR4 on neutrophils to induce autophagy, leading to NET ...

miR‐185‐5p Derived From hUC‐MSC Exosomes via Suspension Culture Under Hypoxic Conditions Promotes Scarless Wound Healing in Mice by Precisely Regulating Collagen I/III Regeneration

open access: yesAdvanced Science, EarlyView.
miR‐185‐5p derived from UC‐MSC exosomes via hypoxic suspension culture promotes scarless wound healing in mice by precisely regulating collagen I/III regeneration. Exosomes derived from UC‐MSCs under a three‐dimensional bioreactor and hypoxic conditions were injected into the skin wounds of mice.

Redox metabolism in cell senescence: focusing on contributions from the metabolomic field

open access: yesFrontiers in Molecular Biosciences
Cell senescence is triggered by stressful stimuli, including telomere attrition, genotoxic agents, and strong mitogenic signals. This state is characterized by proliferation arrest and acquisition of a senescence-associated secretory phenotype. Senescent
