Effects of triggers of senescence and senolysis in murine pancreatic cancer cells

Hepatobiliary & Pancreatic Diseases International
The combination of senescence triggers with senolytic drugs is considered a promising new approach to cancer therapy. Here, we studied the efficacy of the genotoxic agent etoposide (Eto) and irradiation in inducing senescence of Panc02 pancreatic cancer cells, and the capability of the Bcl-2 inhibitor navitoclax (ABT-263; Nav) to trigger senolysis ...

Apoptotic priming in senescence predicts specific senolysis by quantitative analysis of mitochondrial dependencies

Cell Death & Differentiation
Cellular senescence contributes to a variety of pathologies associated with aging and is implicated as a cellular state in which cancer cells can survive treatment. Reported senolytic drug treatments act through varying molecular mechanisms, but heterogeneous efficacy across the diverse contexts of cellular senescence indicates a need for predictive ...

Abstract 5342: Inhibition of glutaminase elicits senolysis in therapy-induced senescent melanoma cells

Cancer Research
Abstract The cyclin D1-Cyclin-Dependent Kinases 4 and 6 (CDK4/6) complex is crucial for the development of melanoma. We previously demonstrated that targeting CDK4/6 using small molecule inhibitors (CDK4/6i) suppresses BrafV600E melanoma growth in vitro and in vivo through induction of cellular senescence.
