Localization of d-serine and serine racemase in neurons and neuroglias in mouse brain

Neurological Sciences, 2010
D-serine is a novel candidate for an intrinsic ligand for the glycine site of N-methyl-D-aspartate (NMDA) receptors in mammalian brain. D-serine and serine racemase, which produces D-serine from L-serine, have long been presumed to be localized in astrocytes. However, we have reported that D-serine immunoreactivity was observed in neurons in rats.

Triple serine loop region regulates the aspartate racemase activity of the serine/aspartate racemase family

Amino Acids, 2017
Recently, we cloned and characterized eleven serine and aspartate racemases (SerR and AspR, respectively) from animals. These SerRs and AspRs are not separated by their racemase functions and form a serine/aspartate racemase family cluster based on phylogenetic analysis.

Occurrence of D-serine in rice and characterization of rice serine racemase

Phytochemistry, 2009
Germinated, unpolished rice was found to contain a substantial amount of D-serine, with the ratio of the D-enantiomer to the L-enantiomer being higher for serine than for other amino acids. The relative amount of D-serine (D/(D+L)%) reached approximately 10% six days after germination. A putative serine racemase gene (serr, clone No.

Preparation and Assay of Recombinant Serine Racemase

2011
Serine racemase is a glial and neuronal enzyme that reversibly converts L-serine to D-serine, an endogenous co-agonist of N-methyl-D-aspartate receptor type glutamate receptors (NMDARs). Here we present methods to recombinantly express and purify serine racemase in bacteria and two complementary ways to determine D-serine levels in unknown samples ...

Serine racemase: an unconventional enzyme for an unconventional transmitter

Amino Acids, 2012
The discovery of large amounts of D-serine in the brain challenged the dogma that only L-amino acids are relevant for eukaryotes. The levels of D-serine in the brain are higher than many L-amino acids and account for as much as one-third of L-serine levels.

Regulation of serine racemase activity by amino acids

Molecular Brain Research, 2005
The effects of various amino acids on the activity of serine racemase, purified from mouse brain, were examined. Those acting as inhibitors included compounds with electron withdrawing groups on the beta-carbon of alanine (beta-halo-alanines and L-serine-O-sulfate), which can act as enzyme-activated inhibitors, and compounds containing beta-SH groups ...

Serine Racemase with Catalytically Active Lysinoalanyl Residue*

The Journal of Biochemistry, 2009
Serine racemase synthesizes d-serine, a physiological agonist of the NMDA receptor in mammalian brains. Schizosaccharomyces pombe produces serine racemase (spSR) that is highly similar to the brain enzyme. Our mass-spectrometric and X-ray studies revealed that spSR is modified with its natural substrate serine. spSR remains partially active even though

Localization and expression of serine racemase in Arabidopsis thaliana

Amino Acids, 2008
Arabidopsis plants transformed by promoter of A. thaliana serine racemase fused with beta-glucuronidase (GUS) reporter gene showed strong GUS staining in elongating and developing cells such as tip regions of primary and lateral roots, developing leaves, and shoot meristems. RT-PCR and digital northern hybridization showed that expression of the serine

Allosteric Regulation of Mouse Brain Serine Racemase

Neurochemical Research, 2002
Serine racemase, purified from mouse brain, consisted of two isoforms. They had similar enzymatic properties and had molecular weights of about 55 kDa based on size exclusion chromatography. This is about twice that reported from its electrophoretic mobility on SDS gels or from the amino acid sequence of the recombinant enzyme.

Schwann cell and epineural fibroblast expression of serine racemase

Brain Research, 2004
D-serine appears to be a natural agonist at the "glycine site" of the NMDA receptor and is created by conversion from L-serine by serine racemase. This racemase has been localized to protoplasmic astrocytes that ensheath synapses and modulate neuronal activity in the CNS, but serine racemase expression in the PNS has not been reported ...
