Functional Mapping of Neurodevelopmental Disease Pathways to Key Neurodevelopmental Processes Represented in the Developmental Neurotoxicity In Vitro Testing Battery

open access: yesAdvanced Science, EarlyView.
Human‐relevant methods are essential for modern chemical safety assessment. This study helps define the capabilities and boundaries of an in vitro testing battery for developmental neurotoxicity by exploring its biological applicability domain. By linking neurodevelopmental disease‐related pathways to key neurodevelopmental processes, the work enhances

Comparative study assessing effects of sonic hedgehog and VEGF in a human co-culture model for bone vascularisation strategies [PDF]

open access: yesEuropean Cells & Materials, 2011
The morphogen sonic hedgehog (Shh) seems to mediate adult repair processes in bone regeneration and vascularisation. In this study we investigated the effects of Shh on co-cultures consisting of human primary osteoblasts and outgrowth endothelial cells ...

Basal Suppression of the Sonic Hedgehog Pathway by the G-Protein-Coupled Receptor Gpr161 Restricts Medulloblastoma Pathogenesis

open access: yesCell Reports, 2018
Summary: Sonic hedgehog (Shh) determines cerebellar granule cell (GC) progenitor proliferation and medulloblastoma pathogenesis. However, the pathways regulating GC progenitors during embryogenesis before Shh production by Purkinje neurons and their ...

An inversion involving the mouse Shh locus results in brachydactyly through dysregulation of Shh expression [PDF]

open access: yesJournal of Clinical Investigation, 2005
Short digits (Dsh) is a radiation-induced mouse mutant. Homozygous mice are characterized by multiple defects strongly resembling those resulting from Sonic hedgehog (Shh) inactivation. Heterozygous mice show a limb reduction phenotype with fusion and shortening of the proximal and middle phalanges in all digits, similar to human brachydactyly type A1,

Shh critically mediates cholangiocyte differentiation.

open access: yes, 2016
A. Reduced Shh dosing reduces mRNA levels of CK7, CK19, and FN (++Shh = 100 ng/ml Shh administered for 8 days, +Shh = 100 ng/ml Shh administered for 4 days, -Shh = no Shh administration). B.

Ecological Adaptation Mechanisms Underlying Successful Plant Reproduction

open access: yesAdvanced Science, EarlyView.
During floral induction, various environmental and endogenous signals converge to regulate the florigen protein, which is transported from leaves to the SAM to initiate flowering. Within the SAM, a complex network of receptor kinases and small peptides orchestrates floral development with high spatiotemporal precision.

Epithelial cells supply Sonic Hedgehog to the perinatal dentate gyrus via transport by platelets

open access: yeseLife, 2015
Dentate neural stem cells produce neurons throughout life in mammals. Sonic hedgehog (Shh) is critical for maintenance of these cells; however, the perinatal source of Shh is enigmatic.

Evasion of cell senescence in SHH medulloblastoma [PDF]

open access: yesCell Cycle, 2016
The mechanisms leading to brain tumor formation are poorly understood. Using Ptch1+/- mice as a medulloblastoma model, sequential mutations were found to shape tumor evolution. Initially, medulloblastoma preneoplastic lesions display loss of heterozygosity of the Ptch1 wild-type allele, an event associated with cell senescence in preneoplasia ...

tBid‐Mediated Genetic Ablation of Connective Tissue Cells Reveals Their Key Regulatory Function During Limb Regeneration in Axolotls

open access: yesAdvanced Science, EarlyView.
We establish a tBid‐mediated cell ablation system in axolotls, achieve rapid and efficient ablation of multiple cell types, including muscle stem cell, spinal cord cell, and connective tissue (CT) cells. We investigate the role of CT using tBid‐mediated CT ablation and identify its essential role for limb development and regeneration.
