The (Sialyl) Tn antigen: Contributions to immunosuppression in gastrointestinal cancers [PDF]

open access: yesFrontiers in Oncology, 2023
Cellular signaling pathways are intricately regulated to maintain homeostasis. During cancer progression, these mechanisms are manipulated to become harmful. O-glycosylation, a crucial post-translational modification, is one such pathway that can lead to

Curcumin-mediated transcriptional regulation of human N-acetylgalactosamine-α2,6-sialyltransferase which synthesizes sialyl-Tn antigen in HCT116 human colon cancer cells [PDF]

open access: yesFrontiers in Molecular Biosciences, 2022
Human N-acetylgalactosamine-α2,6-sialyltransferase (hST6GalNAc I) is the major enzyme involved in the biosynthesis of sialyl-Tn antigen (sTn), which is known to be expressed in more than 80% of human carcinomas and correlated with poor prognosis in ...

Sialyl-Tn Antigen-Imprinted Dual Fluorescent Core-Shell Nanoparticles for Ratiometric Sialyl-Tn Antigen Detection and Dual-Color Labeling of Cancer Cells. [PDF]

open access: yesACS Appl Nano Mater, 2022
Sialyl-Tn (STn or sialyl-Thomsen-nouveau) is a carbohydrate antigen expressed by more than 80% of human carcinomas. We here report a strategy for ratiometric STn detection and dual-color cancer cell labeling, particularly, by molecularly imprinted polymers (MIPs).

Challenges in Antibody Development against Tn and Sialyl-Tn Antigens [PDF]

open access: yesBiomolecules, 2015
The carbohydrate antigens Tn and sialyl-Tn (STn) are expressed in most carcinomas and usually absent in healthy tissues. These antigens have been correlated with cancer progression and poor prognosis, and associated with immunosuppressive ...

Using Synthetic Glycans to Investigate Anti-Glycan Antibodies and Explore Their Medical Potential. [PDF]

open access: yesAngew Chem Int Ed Engl
Anti‐glycan antibodies are essential in health and disease. Access to novel glycan structures paves the way for progress in antibody profiling for biomarker discovery, antibody development, and vaccine design. We summarize the strategies to synthesize and utilize synthetic glycans for the development and application of anti‐glycan antibodies in basic ...

Cosmc loss induces truncated O-glycosylation and accelerates Kras-driven pancreatic carcinogenesis [PDF]

open access: yesNeoplasia: An International Journal for Oncology Research
Aberrant mucin-type O-glycosylation is a common feature of pancreatic ductal adenocarcinoma (PDAC), yet its functional contribution to pancreatic tumorigenesis remains incompletely defined.

Comprehensive Profiling Reveals Sialyl-Tn Upregulation and Prognostic Value in Prostate Cancer. [PDF]

open access: yesPathol Int
This study utilises a novel antibody (L2A5) to comprehensively monitor sialyl‐Tn antigen (sTn) expression levels in clinical prostate cancer tissues. We reveal sTn is expressed in 44% of prostate tumours and find prostate cancer patients with high sTn levels have significantly poorer survival times.

Efficient chemoenzymatic synthesis of sialyl Tn-antigens and derivatives. [PDF]

open access: yesChem Commun (Camb), 2011
An N-terminal and C-terminal truncated recombinant α2-6-sialyltransferase cloned from Photobacterium sp. JH-ISH-224, Psp2,6ST(15-501)-His(6), was shown to be an efficient catalyst for one-pot three-enzyme synthesis of sialyl Tn (STn) antigens and derivatives containing natural and non-natural sialic acid forms.

Phage Display Reveals VLRB-Mediated Recognition of Minimal Tumor Glycan Antigen Sialyl-Tn [PDF]

open access: yesCurrent Issues in Molecular Biology
Sialyl-Tn (sTn) is a tumor-associated carbohydrate antigen (TACA) abundantly expressed by various types of carcinomas. While conventional antibody-based platforms have traditionally been used for the detection and targeting of sTn, alternative binding ...

Protein O-Glycosylation Shapes Lymphatic Endothelia and Lymph Node Macrophage Populations. [PDF]

open access: yesFASEB J
To study the functional roles of O‐glycans on lymphatic endothelia, we deleted Cosmc, the chaperone required for core‐1 O‐glycan synthesis. Loss of core‐1 O‐glycans entailed major changes in glycoprotein composition of lymphatic endothelial cells (LEC), including reduced podoplanin (Pdpn), CD44, and Lyve‐1, and increased ICAM‐1; reduced CD44 was caused
