Molecular, Cellular, and Physiological Characterization of Sirtuin 7 (SIRT7) [PDF]
Sirtuin 7 (SIRT7), a histone 3 lysine 18 (H3K18) deacetylase, functions at chromatin to suppress endoplasmic reticulum (ER) stress and mitochondrial protein folding stress (PFS(mt)), and prevent the development of fatty liver disease and hematopoietic stem cell aging.

Castration‐resistant prostate cancer (CRPC) remains sensitive to ferroptosis, but its intrinsic resistance is poorly understood. Here, we identify NFIB as a master suppressor. SIRT7‐mediated NFIB acetylation drives its liquid–liquid phase separation, which promotes SLC3A2 transcription to inhibit ferroptosis.

Characterization of Sirt2 using conditional RNAi in mice [PDF]
Within the past eight years, RNA interference (RNAi) has emerged as a powerful experimental tool for gene function analysis in mice. Reversible control of shRNA mediated RNAi has been achieved by using a tetracycline (tet)-inducible promoter.

SIRT7-dependent deacetylation of CDK9 activates RNA polymerase II transcription [PDF]
Abstract SIRT7 is an NAD+-dependent protein deacetylase that regulates cell growth and proliferation. Previous studies have shown that SIRT7 is required for RNA polymerase I (Pol I) transcription and pre-rRNA processing. Here, we took a proteomic approach to identify novel molecular targets and characterize the role of SIRT7 in non ...

Cerebral ischemic injury continues to be a leading cause of mortality and disability worldwide. Although Sirtuin 7 (SIRT7), a desuccinylase, is known to regulate protein expression, its role in cerebral ischemia remains unclear.

A muscle‐bone endocrine pathway in aging is revealed in which extracellular vesicles released from atrophic skeletal muscle (Aged‐SKM‐EVs) inhibit bone formation. These EVs deliver miR‐125a‐5p to osteoblasts, thereby suppressing the SIRT7‐Sp7 signaling axis and osteogenic differentiation.

Chronic exposure to simulated space conditions predominantly affects cytoskeleton remodeling and oxidative stress response in mouse fetal fibroblasts [PDF]
Microgravity and cosmic rays as found in space are difficult to recreate on earth. However, ground-based models exist to simulate space flight experiments.

The senescence of macrophages leads to a decrease in the expression of Sirtuin3 (Sirt3), which in turn aggravates the NLRP3 inflammasome activation and IL‐1β secretion, thereby promoting the aging‐related osteoporosis. Sirt3‐enriched apoptotic bodies (ABs‐Sirt3) generated through genetic editing approaches can alleviate skeletal aging by targeting ...
