Integrin α5β1 in pancreatic ductal adenocarcinoma: Tumour‒stroma crosstalk, hypoxia and therapeutic targeting

ITGA5/integrin α5β1 promotes PDAC progression by integrating fibronectin‐dependent adhesion, hypoxia‐associated ECM remodelling, pancreatic stellate cell/cancer‐associated fibroblast activation, desmoplasia and tumour‒stroma crosstalk. These processes contribute to impaired drug delivery and therapeutic resistance.

Interferon‐induced transmembrane (IFITM) proteins at the tumour–immune interface: A four‐axis framework for their context‐dependent roles

Interferon‐induced transmembrane (IFITM) function in cancer is set by four contextual coordinates, not by family membership. Acute interferon input drives major histocompatibility complex class I (MHC‐I)‐linked immunogenicity; chronic input drives programmed death‐ligand 1 (PD‐L1)‐coupled resistance.

ADAMTS4 mediates LPS‐induced cardiac injury and myocardial fibrosis through proteolytic cleavage of TSP1

• ADAMTS4 is induced primarily in cardiac fibroblasts during sepsis. • ADAMTS4 cleaves TSP1 at the 236–246 aa functional region. • TSP1 cleavage activates TGF‐β/Smad and NF‐κB signalling in fibroblasts. • Targeting ADAMTS4 reduces fibroblast activation and cardiac fibrosis.
