Matrigel inhibits elongation and drives endoderm differentiation in aggregates of mouse embryonic stem cells

open access: yesFEBS Open Bio, EarlyView.
Stem cell‐based embryo models (SCBEMs) are valuable to study early developmental milestones. Matrigel, a basement membrane matrix, is a critical substrate used in various SCBEM protocols, but its role in driving stem cell lineage commitment is not clearly defined.

MicroRNA-126 inhibits SOX2 expression and contributes to gastric carcinogenesis. [PDF]

open access: yesPLoS ONE, 2011
BACKGROUND: SRY (sex-determining region Y)-box 2 (SOX2) is a crucial transcription factor for the maintenance of embryonic stem cell pluripotency and the determination of cell fate.

Matrix: a complex amalgam of structures and functions in tumor microenvironment

open access: yesFEBS Open Bio, EarlyView.
The matrix is a dynamic, intricate three‐dimensional mesh of biomolecules with both structural and functional properties. This review deals with the complexity of this ‘molecular amalgam’ in the tumor microenvironment and highlights its importance in the maintenance and evolution of tumors by describing certain matrix biomolecules, such as ...

LIN28B Promotes Cancer Cell Dissemination and Angiogenesis

open access: yesAdvanced Biology, EarlyView.
Children diagnosed with high‐risk neuroblastoma have a 5‐year event‐free survival rate of less than 50% and poor outcomes after recurrence. Deregulation of the LIN28B oncogene can be addressed in these patients. Upregulation of LIN28B is shown to support the metastatic cascade.

Heterozygous mutations in SOX2 may cause idiopathic hypogonadotropic hypogonadism via dominant-negative mechanisms

open access: yesJCI Insight, 2023
Pathogenic SRY-box transcription factor 2 (SOX2) variants typically cause severe ocular defects within a SOX2 disorder spectrum that includes hypogonadotropic hypogonadism.

Evidence That SOX2 Overexpression Is Oncogenic in the Lung

open access: yesPLoS ONE, 2010
SOX2 (Sry-box 2) is required to maintain a variety of stem cells, is overexpressed in some solid tumors, and is expressed in epithelial cells of the lung.We show that SOX2 is overexpressed in human squamous cell lung tumors and some adenocarcinomas.

Bioorthogonal Engineering of Cellular Microenvironments Using Isonitrile Ligations

open access: yesAdvanced Functional Materials, EarlyView.
Highly selective chemistries are required for fabrication and post‐cross–linking modification of cell‐encapsulating hydrogels used in tissue engineering applications. Isonitrile ligation reactions represent a promising class of bioorthogonal chemistries for engineering hydrogel‐based cellular microenvironments. Isonitrile‐based hydrogels are stable and

Mutations within Sox2/SOX2 are associated with abnormalities in the hypothalamo-pituitary-gonadal axis in mice and humans [PDF]

open access: yesJournal of Clinical Investigation, 2006
The transcription factor SOX2 is expressed most notably in the developing CNS and placodes, where it plays critical roles in embryogenesis. Heterozygous de novo mutations in SOX2 have previously been associated with bilateral anophthalmia/microphthalmia, developmental delay, short stature, and male genital tract abnormalities.

Alleviation of Aging‐Related Hallmarks in a Mouse Model of Progeria via a Nanoparticle‐Based Artificial Transcription Factor

open access: yesAdvanced Functional Materials, EarlyView.
Oct4‐nanoscript, a biomimetic nanoparticle‐based artificial transcription factor, precisely regulates cellular rejuvenation by activating Oct4 target genes, restoring epigenetic marks, and reducing DNA damage. In a progeria model, it effectively rescued aging‐associated pathologies and extended lifespan.

The prosensory function of Sox2 in the chicken inner ear relies on the direct regulation of Atoh1. [PDF]

open access: yesPLoS ONE, 2012
The proneural gene Atoh1 is crucial for the development of inner ear hair cells and it requires the function of the transcription factor Sox2 through yet unknown mechanisms. In the present work, we used the chicken embryo and HEK293T cells to explore the
