Squalene epoxidase as the target of antifungal allylamines
Pesticide Science, 1987AbstractThe allylamine clinical antifungals naftifine and terbinafine inhibit ergosterol biosynthesis at the point of squalene epoxidation in a range of fungi pathogenic to man. Filamentous growth is particularly susceptible to this inhibition, cell death occurring at drug concentrations causing only partial inhibition of ergosterol biosynthesis ...

Ellagitannins and Hexahydroxydiphenoyl Esters as Inhibitors of Vertebrate Squalene Epoxidase
Journal of Natural Products, 2001Ellagitannins isolated from various plant sources as well as newly synthesized n-alkyl (C(1)-C(18)) esters of hexahydroxydiphenyl (HHDP) dicarboxylic acid were evaluated as enzyme inhibitors of recombinant rat squalene epoxidase, a rate-limiting enzyme of cholesterol biosynthesis.

Reengineering the Substrate Tunnel to Enhance the Catalytic Efficiency of Squalene Epoxidase.
Journal of Agricultural and Food ChemistrySqualene epoxidase plays a pivotal role in the biosynthesis of ergosterol, its derivatives, and other triterpenoid compounds by catalyzing the transformation of squalene into 2,3-oxidosqualene.

Squalene Epoxidase — Enzymology and Inhibition
1990Squalene occupies a key position in the biosynthetic sequence from acetate to ergosterol (Fig. 1), being the first lipophilic intermediate in the pathway. The conversion of this 30-carbon hydrocarbon to the tetracyclic lanosterol structure was originally thought to occur in one step without intermediates.

Purification of squalene epoxidase from rat liver microsomes
Biochemical and Biophysical Research Communications, 1980Abstract Squalene epoxidase was purified from rat liver microsomes by DEAE-cellulose, alumina C ν gel, hydroxylapatite, CM-Sephadex C-50 and Cibacron Blue Sepharose 4B in the presence of Triton X-100. The specific activity was increased 50 fold with a yield of about 10%.

Terbinafine resistance has become epidemic as an emerging problem in treatment of dermatohpytosis. This could be attributed in part to a point mutation in the squalene epoxidase (SQLE) gene. In this study, point mutations in the SQLE gene were studied in

[Cloning of the Aspergillus fumigatus squalene epoxidase gene].
Beijing da xue xue bao. Yi xue ban = Journal of Peking University. Health sciences, 2008To clone Aspergillus fumigatus squalene epoxidase gene and to further investigate its role in terbinafine resistance. The A.fumigatus genomic DNA library was transformed into pyrG-A. fumigatus strain protoplasts with polyethylene glycol-mediated transformation protocol.
