Immunometabolic Modulation by Low‐Dose Sodium Butyrate: PI3K/Akt/mTOR Signalling Orchestrates Macrophage Polarization to Protect Against Clostridioides difficile Colitis

open access: yesImmunology, EarlyView.
Clostridioides difficile toxins TcdA and TcdB enter intestinal epithelial cells (IECs) via specific receptors, disrupt tight‐junction integrity and activate PI3K/Akt/mTOR signalling, promoting inflammasome assembly and IL‐1β release. Toxin‐induced IEC apoptosis further weakens the epithelial barrier.

A Mouse Model of Airway Disease: Oncostatin M-Induced Pulmonary Eosinophilia, Goblet Cell Hyperplasia, and Airway Hyperresponsiveness Are STAT6 Dependent, and Interstitial Pulmonary Fibrosis Is STAT6 Independent

open access: yes, 2011
Oncostatin M (OSM), a pleiotropic cytokine of the gp130 cytokine family, has been implicated in chronic allergic inflammatory and fibrotic disease states associated with tissue eosinophilia.

Emerging Roles of PSTPIP2 in Autoimmunity: From Mechanism to Therapeutic Implications

open access: yesImmunology, EarlyView.
PSTPIP2 acts as a key brake on myeloid inflammation by suppressing IL‐1β‐driven signalling, macrophage dysregulation and inflammation‐associated bone remodelling. Its emerging roles in CRMO/CMO, rheumatoid arthritis, SAPHO syndrome and bullous pemphigoid support its potential as a biomarker and therapeutic target.

Innovations in Obesity Treatment: Beyond Adipose Tissue Dysfunction

open access: yesObesity Reviews, EarlyView.
Obesity drives chronic inflammation, insulin resistance, type 2 diabetes, and cancer development through adipocyte dysfunction. Addressing this multisystemic disorder requires integrated strategies beyond diet and exercise, such as thermogenesis activation via menthol or capsinoids and appetite control through GLP‐1/GIP agonists and neuromodulation to ...

LANA promotes nuclear localization of STAT6 independent of Y641-phosphorylation.

open access: yes, 2017
(A) Inhibition of LANA blocks nuclear localization of STAT6. Confocal microscopy of endogenous STAT6 (red) and merged in PEL cells BC3 with lentivirus-mediated constitutively knocked down of LANA (shLANA) or luciferase control (shCtrl).

Persisting Region‐Specific Microglia Activation and Synaptic Imbalance During Remission in a DSS‐Induced Colitis Model

open access: yesGlia, Volume 74, Issue 11, November 2026.
Relapsing colitis region‐specific microglial states beyond remission. Cortical microglia remain pro‐inflammatory and synapse‐disruptive, while hippocampal microglia shift toward a reparative program that maintains neuronal circuit stability. ABSTRACT Ulcerative colitis (UC) patients experience cycles of active gut inflammation and remission, with ...

Immune cells in the pathogenesis of abdominal aortic aneurysm: Roles, regulation and targeted interventions

open access: yesClinical and Translational Discovery, Volume 6, Issue 5, October 2026.
This review provides a comprehensive overview of how macrophages, neutrophils, T cells and other immune cells infiltrate the aortic wall, undergo phenotypic shifts and interact with one another in abdominal aortic aneurysm (AAA). Background Abdominal aortic aneurysm (AAA) is a lethal degenerative vascular disease with no approved clinical drugs. Immune

Turning Off STAT6 with a Targeted Degrader

open access: yesJournal of Medicinal Chemistry
While an attractive target for treating a range of human diseases and conditions, signal transducer and activator of transcription 6 (STAT6) has resisted drug discovery efforts for decades. This Viewpoint summarizes the discovery of a highly potent, selective STAT6 degrader with minimal off-target activity.

Bovine milk regulates collagen synthesis via STAT6.

open access: yes, 2015
(a) Human skin fibroblasts were treated with 1, 5, 10 and 20% bovine milk with or without 200 μM leflunomide, an inhibitor of p-STAT6. After 0,5, 2 and 6h cell lysates were examined for p-STAT6 induction by Western blot. (b) Cells were treated with 1, 5,
