Lactate metabolism and protein lactylation in programmed cell death: From novel mechanism to therapeutic strategies in human diseases

open access: yesClinical and Translational Medicine, Volume 16, Issue 10, October 2026.
This review highlights protein lactylation as a metabolic‐epigenetic regulator linking lactate reprogramming to programmed cell death. By modulating histone and non‐histone targets, lactylation reshapes cell fate decisions and represents a promising biomarker and therapeutic target in human diseases.

A Physiologically Based Pharmacokinetic Model for Clobazam and Stiripentol in Adults and Children

open access: yes, 2014
Purpose To develop a physiologically based pharmacokinetic model in adults and children for clobazam, its active metabolite norclobazam and stiripentol and to account for significant clinical interaction that has been reported when clobazam and ...

Relaxation dynamics vs crystallization kinetics in the amorphous state: the case of Stiripentol [PDF]

open access: yes, 2017
With the aim of finding a correlation between the crystallization kinetics and the molecular dynamics of a substance that would allow prediction of its crystallization time as a function of temperature for a given a relaxation time, we have studied ...

Effectiveness and tolerability of fenfluramine in pediatric and adult patients with developmental and epileptic encephalopathies: A multicenter, retrospective, real‐world clinical‐practice study

open access: yesEpilepsia, Volume 67, Issue 9, Page 4536-4550, September 2026.
Abstract Objective Developmental and epileptic encephalopathies (DEEs) are characterized by drug‐resistant seizures and developmental slowing/regression. We examined the efficacy and tolerability of fenfluramine (FFA) in pediatric and adult patients with Lennox–Gastaut syndrome (LGS), Dravet syndrome (DS), and other DEEs.

Intermittent fasting with medium‐chain triglycerides in drug‐resistant epilepsy: A pilot crossover trial

open access: yesEpilepsia, Volume 67, Issue 9, Page 4618-4630, September 2026.
Abstract Objective Ketogenic dietary therapies can reduce seizure frequency in drug‐resistant epilepsy, but adherence to the classical ketogenic diet is often poor. Intermittent fasting supplemented with medium‐chain triglycerides (MCTs) may offer a more feasible and less restrictive alternative.

Audit of use of stiripentol in adults with Dravet syndrome [PDF]

open access: yes, 2016
OBJECTIVES: There are very few data available in the literature on the use of stiripentol in adults with Dravet syndrome (DS). DS cases are increasingly recognized in adulthood, and more children with DS now survive to adulthood. The aim of the study was

SUDEP and mortality in developmental and epileptic encephalopathies: A meta‐analysis of randomized clinical trials and extension studies

open access: yesEpilepsia, Volume 67, Issue 9, Page 4631-4648, September 2026.
Abstract Objective Developmental and epileptic encephalopathies (DEEs) are associated with high premature mortality and increased risk of sudden unexpected death in epilepsy (SUDEP). However, epidemiological data remain limited, particularly for specific syndromes such as Dravet syndrome (DS), Lennox–Gastaut syndrome (LGS), and infantile epileptic ...

Understanding Postmarketing Requirements and Commitments for CYP3A‐Mediated Drug–Drug Interactions: An Analysis of FDA‐Approved Drugs From 2015 to 2024

open access: yesClinical and Translational Science, Volume 19, Issue 9, September 2026.
ABSTRACT Addressing knowledge gaps at drug approval through drug–drug interaction (DDI)‐related postmarketing requirements (PMRs) and commitments (PMCs) is essential for safe and effective use. This analysis reviewed CYP3A‐related PMRs and PMCs, the most evaluated enzyme in postmarketing studies, to characterize their scope.

(E)-1-(Benzo[d][1,3]dioxol-5-yl)-5,6,6-trimethylhept-4-en-3-one

open access: yesMolbank
The title compound (1) was obtained within a project to synthesize analogs of the antiepileptic drug stiripentol. Compound 1 was synthesized by aldol addition of the lithium enolate of 4-(benzo[d][1,3]dioxol-5-yl)butan-2-one (2) to 3,3-dimethylbutan-2 ...

Stiripentol as Treatment for Epileptic Seizures [PDF]

open access: yes
I denne oppgaven skal stiripentols mekanismer mot epilepsi diskuteres. Stiripentol er et legemiddel mest brukt mot Dravet syndrom, en sjelden barneepilepsi med høy dødsrate.
