Sulfiredoxin: a potential therapeutic agent? [PDF]

open access: yesBiomedicine and Pharmacotherapy, 2005
The importance of antioxidants in maintaining homeostasis has long been accepted and includes antioxidant proteins such as, peroxiredoxin (Prx), superoxide dismutase and glutathione S transferases. Sulfiredoxin (Srx) is a recently identified antioxidant protein with a role in signaling through catalytic reduction of oxidative modifications.

Transcriptional regulation of the AP-1 and Nrf2 target gene sulfiredoxin [PDF]

open access: yesMolecules and Cells, 2009
"Two-cysteine" peroxiredoxins are antioxidant enzymes that exert a cytoprotective effect in many models of oxidative stress. However, under highly oxidizing conditions they can be inactivated through hyperoxidation of their peroxidatic active site cysteine residue. Sulfiredoxin can reverse this hyperoxidation, thus reactivating peroxiredoxins.

Elucidation of the inhibition mechanism of sulfiredoxin using molecular modeling and development of its inhibitors.

Journal of Molecular Graphics and Modelling, 2019
When intracellular reactive oxygen species (ROS) increase, cancer cells are more vulnerable to oxidative stress compared to normal cells; thus, the collapse of redox homeostasis can lead to selective death of cancer cells.

Reversal of 2-Cys peroxiredoxin oligomerization by sulfiredoxin

Biochemical and Biophysical Research Communications, 2013
Hydrogen peroxide (H(2)O(2)) regulates the structure and function of 2-Cys peroxiredoxins (Prxs). Upon oxidation by excess H(2)O(2), Prxs become overoxidized to a sulfinic acid of its peroxidatic cysteine residue, resulting in a structural change from a small oligomer with peroxidase function to a large oligomer with chaperone function.

Comparative analysis of sulfiredoxin and total oxidative stress levels in diabetic individuals with periodontitis-a case-control study.

The Journal of Periodontology, 2022
BACKGROUND The delicate balance between oxidative stress and its antioxidant system can be disrupted in diabetes mellitus (DM) making the tissue susceptible to injury.

Structure-based analysis and rational design of human peroxiredoxin-1's C-terminus-derived peptides to target sulfiredoxin-1 in pancreatic cancer.

Biophysical Chemistry, 2022
Human peroxiredoxin (PRX) family of antioxidant enzymes reduces hydrogen peroxide and alkyl hydroperoxide involved in the redox signaling, among which the widely documented PRX1 is a versatile molecule regulating cell growth, differentiation and ...

Sulfiredoxin-1 protects retinal ganglion cells from high glucose-induced oxidative stress and inflammatory injury by potentiating Nrf2 signaling via the Akt/GSK-3β pathway.

International Immunopharmacology, 2021
Sulfiredoxin-1 (Srxn1) has been acknowledged as a remarkable pro-survival factor in the protection of cells against stress-induced damage. The persistent exposure of retinal ganglion cells (RGCs) to high glucose (HG) in diabetes induces cellular damage ...

The Sulfiredoxin‐Peroxiredoxin axis promotes urethane‐induced lung adenocarcinoma through the regulation of the tumor microenvironment

The FASEB Journal, 2022
Lung adenocarcinoma is the most common type of lung cancer and has a strong association with tobacco smoking. Smoking leads to the accumulation of reactive oxygen species (ROS) and oxidative stress damages to macromolecules, leading to abnormal cell ...
