Low SLC8A1 in Colorectal Cancer Dictates Radiotherapy Resistance Through Calcium‐Dependent ANXA2+sEVs Driving M2 Polarization of TAMs

open access: yesAdvanced Science, EarlyView.
SLC8A1 deficiency in colorectal cancer cells increases intracellular calcium signaling, promoting the secretion of ANXA2‐enriched small extracellular vesicles. These vesicles reprogram tumor‐associated macrophages towards an M2 phenotype, which in turn fosters a tumor microenvironment conducive to radioresistance and is associated with poor patient ...

Multi‐omics profiling identifies TNFRSF18 as a novel marker of exhausted CD8⁺ T cells and reveals tumour‐immune dynamics in colorectal cancer

open access: yesClinical and Translational Medicine
Background Colorectal cancer (CRC) ranks among the most prevalent malignant tumours of the digestive system globally and is associated with unfavourable survival outcomes. The exhaustion of CD8⁺ T cells serves a crucial role in facilitating tumour immune

NFAT1 regulates CD4+ t cell exhaustion

open access: yes, 2016
Nuclear factor of activated T-cells 1 (NFAT1) is a transcription factor that is involved in both T cell activation as well as T cell anergy and drives distinct context-dependent transcriptional programs.

Autophagy Orchestrates Anti‐Tumor Immunity to Enhance Chemosensitivity via the FBXW2/C/EBPβ/TIMP‐2 Axis in Colorectal Cancer

open access: yesAdvanced Science, EarlyView.
Chemotherapy leverages tumor‐cell autophagy to reshape the colorectal cancer immune microenvironment. Autophagy degrades FBXW2 to promote C/EBPβ‐mediated TIMP‐2 transcription and MMP‐9 suppression, driving intra‐tumoral CD8+ T‐cell infiltration. Targeting MMP‐2/9 or restoring TIMP‐2 overcomes chemo‐resistance in autophagy‐deficient tumors. ABSTRACT The

Intrasplenic Thymus Organogenesis from Injectable Tissue Fragments Restores Functional T‐Cell Immunity

open access: yesAdvanced Science, EarlyView.
Clinical intramuscular thymus transplantation yields only short‐lived efficacy and marginal therapeutic benefits. Benefiting from the spleen's intrinsic strengths—rapid vascular perfusion, abundant developmental factors, and resident progenitors—the intrasplenic thymic grafts achieve robust thymic regeneration and substantial T‐cell reconstitution ...

ZBTB18 Dysfunction Promotes Neuropathic Pain via CHD4‐based Epigenetic Disinhibition of CLIC1 Channels in Sensory Neurons

open access: yesAdvanced Science, EarlyView.
In this study, we identify a novel functional role of ZBTB18 in regulating trigeminal‐mediated neuropathic pain. Nerve injury reduces ZBTB18 in trigeminal ganglion neurons, impairing CHD4/NuRD recruitment and de‐repressing Clic1. Elevated CLIC1 enhances chloride channel activity and neuronal hyperexcitability, thereby driving pain.
