Neoantigen cancer vaccines: a new star on the horizon

open access: yesCancer Biology & Medicine
Immunotherapy represents a promising strategy for cancer treatment that utilizes immune cells or drugs to activate the patient’s own immune system and eliminate cancer cells.

Signalling initiated with CD4–TCR or TCR–TCR interactions: comparison of tyrosine phosphorylation patterns and CD45 effects

open access: yesImmunology Letters, 1998
Antigen-triggered response in T cells is mediated by the T cell receptor (TCR)/CD3-complex. This signalling, however, is modulated by a number of other surface molecules. Among the most important of these is the CD4/CD8 molecule which associates with the TCR/CD3-complex and binds to the MHC complex.

Tailoring Vascular‐Immune Homeostasis via Manganese‐DNA Complex‐Armed Immunogenic Extracellular Vesicles for Pancreatic Cancer Immunotherapy

open access: yesAdvanced Science, EarlyView.
This study demonstrates that Mn2⁺–tumor DNA complexes encapsulated in dendritic cell (DC)– derived immunogenic extracellular vesicles (EVDC@Mn‐DNA) act as a DC‐specific cGAS– STING activator. EVDC@Mn‐DNA treatment enhances intratumoral DC activation, improves tumor vascular function, promotes CD8⁺ T cell activity, and suppresses pancreatic tumor growth,

TCR Convergence in Individuals Treated With Immune Checkpoint Inhibition for Cancer. [PDF]

open access: yes, 2019
Tumor antigen-driven selection may expand T cells having T cell receptors (TCRs) of shared antigen specificity but different amino acid or nucleotide sequence in a process known as TCR convergence. Substitution sequencing errors introduced by TCRβ (TCRB)

Anti‐PD‐1 Nanobody‐Armored MSLN CAR‐T Therapy for Malignant Mesothelioma: Preclinical and Clinical Studies

open access: yesAdvanced Science, EarlyView.
A novel therapy using engineered immune cells (NAC‐T cells) showed promise for refractory malignant mesothelioma. Based on the encouraging preclinical data, the first‐in‐human trial is initiated, demonstrating tolerable safety and promising anti‐tumor activity (ORR 63.6%, DCR 100%, including one CR).

TCR signaling emerges from the sum of many parts.

open access: yesFrontiers in Immunology, 2012
How does T cell receptor signaling begin? Answering this question requires an understanding of how the parts of the molecular machinery that mediates this process fit and work together. Ultimately this molecular architecture must (i) trigger the relay of

Western Diet Inhibits FUT7‐Mediated Treg Intestinal Homing to Disrupt the Homeostasis of Intestinal Epithelial Cells in Crohn's Disease

open access: yesAdvanced Science, EarlyView.
In this study, we constructed a nanoparticle that was CD4 antibody‐mediated targeted, Fut7‐expressing plasmid‐loaded cationic liposome, namely CD4‐LDP‐Fut7. Upregulation of Fut7 expression in Tregs by using CD4‐LDP‐Fut7 can increase Treg homing to the intestine, thereby facilitating repair of the intestinal epithelial barrier and inhibiting ...

Enzymatic Remodelling of Tumour Microenvironment Enhances Anti‐CEACAM5 CAR T‐Cell Efficacy Against Colorectal Cancer

open access: yesAdvanced Science, EarlyView.
This study shows anti‐CEACAM5 CAR T‐cells are ineffective against colorectal cancer (CRC) because of CEACAM5 sequestration at intercellular junctions and the thick tumour cell glycocalyx. Enzymatic treatments of CRC cell monolayer/tissue section with trypsin or hyaluronidase restore CEACAM5 availability, enhance CAR T‐cell activation, increase ...
