Thiopurine Methyltransferase and Thiopurine Metabolite Testing in Patients with Inflammatory Bowel Disease who are Taking Thiopurine Drugs

Pharmacogenomics, 2009
Thiopurine methyltransferase genotyping and thiopurine metabolite testing has been established as an adjunct to monitoring patients taking thiopurine drugs. This special report describes the clinical implications for this type of testing for patients with inflammatory bowel disease who are taking thiopurine drugs.

The Effect of Thiopurine Methyltransferase Expression on Sensitivity to Thiopurine Drugs

Molecular Pharmacology, 2002
Although the thiopurine drugs 6-mercaptopurine (6-MP) and 6-thioguanine (6-TG) are well established agents for the treatment of leukemia, controversies remain regarding their main mode of action. Previous evidence has suggested that although 6-TG exerts a cytotoxic effect through incorporation of 6-thioguanine nucleotides into newly synthesized DNA ...

Pharmacogenetics of Thiopurine S-Methyltransferase and Thiopurine Therapy

Therapeutic Drug Monitoring, 2004
Most medications exhibit wide interpatient variability in their efficacy and toxicity. For many medications, these interindividual differences result in part from polymorphisms in genes encoding drug-metabolizing enzymes, drug transporters, and/or drug targets (eg, receptors, enzymes).

Low-dose thiopurine with allopurinol co-therapy overcomes thiopurine intolerance and allows thiopurine continuation in inflammatory bowel disease

Digestive and Liver Disease, 2018
To assess the utility and tolerability of thiopurine-allopurinol co-therapy in inflammatory bowel disease (IBD) patients with intolerance to thiopurine monotherapy.A retrospective observational study assessed cases of thiopurine intolerance then switched to thiopurine allopurinol co-therapy between 2011 and 2015 at two centres.

Differential effects of mesalazine formulations on thiopurine metabolism through thiopurine S‐methyltransferase inhibition

Journal of Gastroenterology and Hepatology, 2021
AbstractBackground and AimThiopurines are often used in combination with mesalazine for the treatment of ulcerative colitis (UC). Mesalazine formulations are delivered to the digestive tract by various delivery systems and absorbed as 5‐aminosalicylic acid (5‐ASA).

The Future of Thiopurine Pharmacogenomics

Pharmacogenomics, 2012
Since their invention [1], thiopurines (azathioprine, 6-mercaptopurine [6MP], and 6-thioguanine [6TG]) have significantly advanced the treatment of acute lymphoblastic leukemia, organ transplantation, inflammatory bowel disease (IBD) and various autoimmune diseases.

Reducing risk in thiopurine therapy

Xenobiotica, 2019
The thiopurine drugs azathioprine and mercaptopurine are effective in the treatment of disorders of immune regulation and acute lymphoblastic leukaemia. Although developed in the 1950s, thiopurines remained relevant in the anti-tumour necrosis factor biologic era, finding widespread use as a co-immunomodulator.

Are Thiopurines Always Contraindicated After Thiopurine‐Induced Pancreatitis in Inflammatory Bowel Disease?

Journal of Pediatric Gastroenterology and Nutrition, 2013
ABSTRACT Background and Aims: Thiopurine use in inflammatory bowel disease (IBD) is well established for maintenance of disease remission. Approximately 3% of patients with IBD develop thiopurine‐induced pancreatitis (TIP) as an idiosyncratic reaction.

Improved HPLC methodology for monitoring thiopurine metabolites in patients on thiopurine therapy

Clinical Biochemistry, 2004
Standardization of thiopurine metabolite testing is currently lacking. This paper presents in-depth methodological analysis and optimization of two currently available HPLC procedures (Lennard-Singleton [J. Chromatogr. 583 (1992) 83] and Dervieux-Boulieu [Clin. Chem.

Role of Thiopurine Metabolite Testing and Thiopurine Methyltransferase Determination in Pediatric IBD

Journal of Pediatric Gastroenterology and Nutrition, 2013
ABSTRACTThiopurines have been used in inflammatory bowel disease (IBD) for >30 years, and measurements of both thiopurine methyltransferase (TPMT) and thiopurine (TP) metabolites, 6‐thioguanine nucleotides (6‐TGN) and 6‐methylmercaptopurine (6‐MMP), have been readily available.
