AN INSIGHT INTO MEDICINAL PROPERTIES OF QUINOXALINE SCAFFOLD, WITH A FOCUS ON THEIR ANTICANCER POTENTIAL AS INTERCALATING AGENTS [PDF]

open access: yesAl-Azhar Journal of Pharmaceutical Sciences
The antitumor properties of synthetic heterocyclic compounds are among the most important properties that can be made use in medicinal chemistry. More specifically, their significant cytotoxic effects against many types of human tumor cells, as well as ...

Topoisomerases in kinetoplastids

open access: yesParasitology Today, 1996
Topoisomerases are enzymes that mediate topological changes in DNA that are essential for nucleic acid biosynthesis and for cell survival. The kinetoplastid protozoa, which include pathogenic trypanosomes and Leishmania, have yielded an interesting variety of purified topoisomerase activities as well as several topoisomerase genes.

AFM studies of DNA structures extracted from adriamycin treated and non-treated Ehrlich tumor cells [PDF]

open access: yesJournal of the Serbian Chemical Society, 2005
Atomic force microscopy (AFM), a unique tool to investigate drug treatment of cancer cells, was used to analyze the anti-neoplastic activity of adriamycin by comparing DNA structures of non-treated and adriamycin-treated Ehrlich tumor cells.

Cracking the Cytotoxicity Code: Apoptotic Induction of 10-Acetylirciformonin B is Mediated through ROS Generation and Mitochondrial Dysfunction

open access: yesMarine Drugs, 2014
A marine furanoterpenoid derivative, 10-acetylirciformonin B (10AB), was found to inhibit the proliferation of leukemia, hepatoma, and colon cancer cell lines, with selective and significant potency against leukemia cells.

Variation of Structure and Cellular Functions of Type IA Topoisomerases across the Tree of Life

open access: yesCells
Topoisomerases regulate the topological state of cellular genomes to prevent impediments to vital cellular processes, including replication and transcription from suboptimal supercoiling of double-stranded DNA, and to untangle topological barriers ...

The lignan niranthin poisons Leishmania donovani topoisomerase IB and favours a Th1 immune response in mice

open access: yesEMBO Molecular Medicine, 2012
Niranthin, a lignan isolated from the aerial parts of the plant Phyllanthus amarus, exhibits a wide spectrum of pharmacological activities. In the present study, we have shown for the first time that niranthin is a potent anti‐leishmanial agent.

Type IA Topoisomerases as Targets for Infectious Disease Treatments

open access: yesMicroorganisms, 2021
Infectious diseases are one of the main causes of death all over the world, with antimicrobial resistance presenting a great challenge. New antibiotics need to be developed to provide therapeutic treatment options, requiring novel drug targets to be ...

MITF maintains genome stability in nonmelanocyte lineages

open access: yesMolecular Oncology, EarlyView.
MITF is essential for melanocyte survival and acts as an oncogene in 10%–20% of melanomas. We show that MITF depletion causes genome instability in nonmelanocytic cells, leading to LATS2‐mediated P53 activation, cell cycle arrest, and apoptosis. This study highlights the role of MITF as a genome maintenance factor beyond the melanocyte lineage. Created

Neolignans from Licaria chrysophylla and Licaria aurea with DNA topoisomerase II-α inhibitory activity

open access: yesQuímica Nova, 2012
Seven natural neolignans isolated from Licaria chrysophylla and Licaria aurea along with five semisynthetic derivatives were tested for their inhibitory action on DNA-topoisomerase by relaxation assays on pBR322 plasmid DNA.
