Precision Nanomedicine for Traumatic Neural Injury: Functional Paradigms and Translational Frameworks [PDF]
This review presents a pathology‐driven framework unifying nanomedicine across TBI, SCI, and PNI. Epidemiological evidence and comparative single‐cell transcriptomic reanalysis define shared pathological modules, injury‐specific effectors, and therapeutic bottlenecks. Six functional paradigms connect nanomaterial structure and biological fate to injury‐

TPBE scaffolds, in conjunction with mild photothermal effects, effectively activate the PI3K/Akt signaling cascade, thereby promoting early neuronal differentiation and neurite growth. Short‐term (12 W) in vivo implantation studies revealed no significant pathological damage to major organs, providing preliminary evidence of the material's excellent ...

Pathological tissue rigidity mechanoprimes microglia by enhancing actin cytoskeleton–nucleus coupling and chromatin opening at rigidity‐responsive cis‐regulatory elements (mechanoCREs). Subsequent NF‐κB/p65 signaling converges on this permissive regulatory state to amplify inflammatory gene expression and microglial activation.

PLXDC2 siRNA‐Mediated Intervention Attenuates Microglial Senescence Through cGAS‐STING Signaling
PLXDC2 was identified as a microglial target in Parkinson's disease through human and mouse single‐cell analyses. Ferritin‐armed biomimetic nanovesicles delivered PLXDC2 siRNA, deferoxamine, and CeO2 nanoparticles across the blood–brain barrier, reducing cGAS‐STING activation, iron burden, oxidative stress, and senescence in microglia, thereby ...

Calpain Inhibition Reduces Axolemmal Leakage in Traumatic Axonal Injury
Calcium-induced, calpain-mediated proteolysis (CMSP) has recently been implicated to the pathogenesis of diffuse (traumatic) axonal injury (TAI). Some studies suggested that subaxolemmal CMSP may contribute to axolemmal permeability (AP) alterations ...

Adult human organotypic cortical slices undergo marked neuronal loss, laminar disruption, and glial activation, yet their network activity progressively stabilizes after a variable period. Electrophysiology, longitudinal calcium imaging, and quantitative histology show that structurally remodeled human circuits show adaptive capacity resulting in ...

Axonal integrity predicts cortical reorganisation following cervical injury [PDF]
Traumatic spinal cord injury (SCI) leads to disruption of axonal architecture and macroscopic tissue loss with impaired information flow between the brain and spinal cord-the presumed basis of ensuing clinical ...

A Gut‐Brain Axis Underlying Succinate‐Associated Compulsive‐Like Behavior
Gut‐derived succinate is associated with an excitatory‐inhibitory imbalance in dmPFC glutamatergic neurons, involving microglial SUCNR1 activation and vagal afferent signaling via upregulated gut 5‐HT4 receptors along the DVC‐CeA‐dmPFC circuit, changes that are linked to excessive compulsive‐like behavior.

Development of a paediatric model of diffuse traumatic brain injury in ferrets
Traumatic brain injury (TBI) is a leading cause of death and disability in children, especially those under five, with younger children more vulnerable to persistent cognitive and neuropsychological effects due to disrupted brain development.
