Targeted activity of the small molecule kinase inhibitor Pz-1 towards RET and TRK kinases

open access: yesScientific Reports, 2021
We have recently described Pz-1, a benzimidazole-based type-2 RET and VEGFR2 inhibitor. Based on a kinome scan, here we show that Pz-1 is also a potent (IC50

NGF causes TrkA to specifically attract microtubules to lipid rafts. [PDF]

open access: yesPLoS ONE, 2012
Membrane protein sorting is mediated by interactions between proteins and lipids. One mechanism that contributes to sorting involves patches of lipids, termed lipid rafts, which are different from their surroundings in lipid and protein composition ...

Functional interaction between p75NTR and TrkA: the endocytic trafficking of p75NTR is driven by TrkA and regulates TrkA-mediated signalling [PDF]

open access: yesBiochemical Journal, 2004
The topology and trafficking of receptors play a key role in their signalling capability. Indeed, receptor function is related to the microenvironment inside the cell, where specific signalling molecules are compartmentalized. The response to NGF (nerve growth factor) is strongly dependent on the trafficking of its receptor, TrkA.

C1GALT1 expression predicts a favorable prognosis and suppresses malignant phenotypes via TrkA signaling in neuroblastoma

open access: yesOncogenesis, 2022
Neuroblastoma (NB) is a childhood tumor derived from the sympathoadrenal lineage of the neural crest progenitor cells. Core 1 β1,3-galactosyltransferase (C1GALT1) controls the crucial step of GalNAc-type O-glycosylation, and its altered expression ...

GIPC Is Recruited by APPL to Peripheral TrkA Endosomes and Regulates TrkA Trafficking and Signaling [PDF]

open access: yesMolecular and Cellular Biology, 2006
GIPC is a PDZ protein located on peripheral endosomes that binds to the juxtamembrane region of the TrkA nerve growth factor (NGF) receptor and has been implicated in NGF signaling. We establish here that endogenous GIPC binds to the C terminus of APPL, a Rab5 binding protein, which is a marker for signaling endosomes.

Direct interaction of TrkA/CD44v3 is essential for NGF-promoted aggressiveness of breast cancer cells

open access: yesJournal of Experimental & Clinical Cancer Research, 2022
Background CD44 is a multifunctional membrane glycoprotein. Through its heparan sulfate chain, CD44 presents growth factors to their receptors. We have shown that CD44 and Tropomyosin kinase A (TrkA) form a complex following nerve growth factor (NGF ...

Non-TrkA-expressing small DRG neurons are lost in TrkA deficient mice [PDF]

open access: yesThe Journal of Neuroscience, 1995
Experiments over the past decade in which NGF/TrkA signaling has been abolished by antibodies or targeted gene mutations have shown that 70- 85% of dorsal root ganglion (DRG) neurons require NGF for survival during development. There is consensus that many of the NGF-dependent neurons are small-diameter, peptidergic neurons subserving nociception ...

Novel mutations in UL24 and gH rescue efficient infection of an HSV vector retargeted to TrkA

open access: yesMolecular Therapy: Methods & Clinical Development, 2023
Transductional targeting of herpes simplex virus (HSV)-based gene therapy vectors offers the potential for improved tissue-specific delivery and can be achieved by modification of the viral entry machinery to incorporate ligands that bind the desired ...

Correlation of Nerve Growth Factor and Its Receptors Expression with Perineural Invasion in Early-stage Cervical Cancer

open access: yesZhongliu Fangzhi Yanjiu, 2018
Objective To evaluate the potential association of nerve growth factor (NGF) and its receptors TrkA and p75NRT with perineural invasion(PNI) and other clinicopathological features of early-stage cervical cancer.

Multivesicular bodies mediate long-range retrograde NGF-TrkA signaling

open access: yeseLife, 2018
The development of neurons in the peripheral nervous system is dependent on target-derived, long-range retrograde neurotrophic factor signals. The prevailing view is that target-derived nerve growth factor (NGF), the prototypical neurotrophin, and its ...
