Ca2+-induced PRE-NMR changes in the troponin complex reveal the possessive nature of the cardiac isoform for its regulatory switch. [PDF]

open access: yesPLoS ONE, 2014
The interaction between myosin and actin in cardiac muscle, modulated by the calcium (Ca2+) sensor Troponin complex (Tn), is a complex process which is yet to be fully resolved at the molecular level. Our understanding of how the binding of Ca2+ triggers

Development of a specific troponin I detection system with enhanced immune sensitivity using a single monoclonal antibody [PDF]

open access: yesRoyal Society Open Science, 2020
Using an immunoassay in combination with surface plasmon fluorescence spectroscopy (SPFS), we report the rapid detection of troponin I, a valuable biomarker for diagnosis of myocardial infarction.

Computational Studies of Cardiac and Skeletal Troponin

open access: yesFrontiers in Molecular Biosciences, 2019
Troponin is a key regulatory protein in muscle contraction, consisting of three subunits troponin C (TnC), troponin I (TnI), and troponin T (TnT). Calcium association to TnC initiates contraction by causing a series of dynamic and conformational changes ...

Comparison of ARCHITECT chemiluminiscent microparticle immunoassay for determination of Troponin I in serum with AXYM MEIA technology

open access: yesJournal of Health Sciences, 2011
Introduction: The aim of this study was determination of troponin I at serum using Architect (Abbott) and AxSYM System (Abbott). Troponin is regulatory subunit of the troponin complex associate with actin filament within muscle cells and it is a marker ...

Distance distributions and anisotropy decays of troponin C and its complex with troponin I [PDF]

open access: yesBiochemistry, 1991
We used frequency domain measurements of fluorescence resonance energy transfer to recover the distribution of distances between Met 25 and Cys 98 in rabbit skeletal troponin C. These residues were labeled with dansylaziridine as energy donor and 5-(iodoacetamido)eosin as acceptor and are located on the N- and C-terminal lobes of the two-domain protein,

A Case Series of Disproportionate Elevations of Cardiac Troponin and Macrotroponin in Fabry Disease [PDF]

open access: yesJournal of Inborn Errors of Metabolism and Screening
Fabry disease is a rare X-linked lysosomal storage disorder that causes progressive cellular accumulation of glycosphingolipids, leading to various end-organ manifestations such as chronic kidney disease and cardiomyopathy.

Effects of troponin T cardiomyopathy mutations on the calcium sensitivity of the regulated thin filament and the actomyosin cross-bridge kinetics of human β-cardiac myosin. [PDF]

open access: yesPLoS ONE, 2013
Hypertrophic cardiomyopathy (HCM) and dilated cardiomyopathy (DCM) lead to significant cardiovascular morbidity and mortality worldwide. Mutations in the genes encoding the sarcomere, the force-generating unit in the cardiomyocyte, cause familial forms ...

The calcium‐induced switch in the troponin complex probed by fluorescent mutants of troponin I [PDF]

open access: yesEuropean Journal of Biochemistry, 2003
The Ca2+‐induced transition in the troponin complex (Tn) regulates vertebrate striated muscle contraction. Tn was reconstituted with recombinant forms of troponin I (TnI) containing a single intrinsic 5‐hydroxytryptophan (5HW). Fluorescence analysis of these mutants of TnI demonstrate that the regions in TnI that respond to Ca2+ binding to the ...

Bovine cardiac troponin subunits: binary complexes and reconstitution of whole troponin

open access: yesFEBS Letters, 1982
The reconstitution of bovine cardiac troponin from its subunits has been investigated using hydrodynamic techniques. Gel filtration (Sephacryl S‐300) and sedimentation velocity experiments indicate that troponin‐C and troponin‐I from a stable binary complex (1:1 mole ratio) with an apparent Stokes' radius of 36 Å (frictional ratio = 1.6).

Calcium induced regulation of skeletal troponin--computational insights from molecular dynamics simulations. [PDF]

open access: yesPLoS ONE, 2013
The interaction between calcium and the regulatory site(s) of striated muscle regulatory protein troponin switches on and off muscle contraction. In skeletal troponin binding of calcium to sites I and II of the TnC subunit results in a set of structural ...
