An investigation into the role and regulation of uncoupling protein 1 (UCP 1)

open access: yes, 2011
The use of the uncoupling protein inhibitor GDP was used to investigate UCP 1 dependent ROS production in BAT and thymus mitochondria. The effect of UCP 1 inhibition by GDP was then corroborated using UCP 1 ablated mice and their wild-type controls. It was shown that UCP 1 was capable of attenuating ROS production in isolated BAT mitochondria.

VDAC1‐Targeted NHK1 Peptide Recovers Mitochondrial Dysfunction Counteracting Amyloid‐β Oligomers Toxicity in Alzheimer's Disease

open access: yesAging Cell, EarlyView.
Amyloid‐β (Aβ) oligomers affect VDAC1 electrophysiology and the physiological metabolic cross‐talk between cytosol and mitochondria. The use of a small, synthetic VDAC1‐antagonist peptide, derived from hexokinase, restores mitochondrial functionality, and respiration in cellular models of Alzheimer's disease. ABSTRACT Mitochondrial dysfunction has been

Age‐Associated Decline in Autophagy Pathways in the Retinal Pigment Epithelium and Protective Effects of Topical Trehalose in Light‐Induced Outer Retinal Degeneration in Mice

open access: yesAging Cell, EarlyView.
Transcriptomic analyses reveal an age‐related decline in autophagy pathways in RPE/choroid. These pathways remain unchanged in early AMD but become bidirectionally dysregulated in later stages, along with increased RPE loss. Pre‐treatment with trehalose eyedrops enhances RPE autophagy flux and prevents light‐induced outer retinal degeneration in mice ...

ElixirSeeker: A Machine Learning Framework Utilizing Fusion Molecular Fingerprints for the Discovery of Lifespan‐Extending Compounds

open access: yesAging Cell, EarlyView.
ElixirSeeker is a machine learning architecture based on a phenotype‐driven drug discovery approach to screen lifespan‐extending compounds. By integrating molecular fingerprints, ElixirSeeker could maximize feature capture of lifespan‐extending compounds.

Brown and beige adipose tissue: New therapeutic targets for metabolic disorders

open access: yesHealth Sciences Review
Brown adipocytes constitute a specialized tissue in heat build-up (i.e., thermogenesis) due to their mitochondrial uncoupling capacity, as they express thermogenic genes, playing a role in the energy metabolism of the whole body in mammals through non ...
