Cytotoxic withanolide constituents of Physalis longifolia [PDF]

open access: yesPlanta Medica, 2011
Fourteen new withanolides, 1-14, named withalongolides A-N, respectively, were isolated from the aerial parts of Physalis longifolia together with eight known compounds (15-22). The structures of compounds 1-14 were elucidated through spectroscopic techniques and chemical methods.

Effect of the withanolides on ER-positive human breast cancer cells.

open access: yes, 2013
(A) Withanolides reduce the viability of MCF-7 cells. MCF-7 cells were treated with the indicated concentrations of WA, HW, AA, or PH for 48 h, and cell viability was determined by the MTT assay.

In silico evaluation of pharmacokinetics properties of withanolides and simulation of their biological activities against Alzheimer’s disease

open access: yes, 2023
The withanolides are naturally occurring steroidal lactones found mainly in plants of the Solanaceae family. The subtribe Withaninae includes species like Withania sominifera, which are a source of many bioactive withanolides.

Evaluation of Withaferin A Content in Withania somnifera Using NMR and HPLC: A Comparative Assessment of Traditional and PCSL Extraction Methods

open access: yesChemistry &Biodiversity, Volume 23, Issue 6, June 2026.
A comparative evaluation of traditional aqueous extractions and innovative pressurised cyclic solid–liquid extraction is presented for Withania somnifera leaves. Quantitative NMR, validated by HPLC, reveals a markedly enhanced preservation and recovery of withaferin A using PCSL.

Withanolides from Physalis coztomatl

open access: yes, 2020
a b s t r a c t Six withanolides (1-6), as well as two known withanolides (physachenolide D 7 and withanoside VI 8), were isolated from the aerial parts of Physalis coztomatl (Solanaceae).

Withanolides from dietary tomatillo suppress HT1080 cancer cell growth by targeting mutant IDH1

open access: yes, 2021
Isocitrate dehydrogenase (IDH) is one key rate-limiting enzyme in the tricarboxylic acid cycle, which is related to various cancers. Tomatillo (Physalis ixocarpa), a special tomato, is widely consumed as nutritious vegetable in Mexico, USA, etc.

Molecular dynamics simulations and experimental studies reveal differential permeability of withaferin-A and withanone across the model cell membrane

open access: yesScientific Reports, 2021
Poor bioavailability due to the inability to cross the cell membrane is one of the major reasons for the failure of a drug in clinical trials. We have used molecular dynamics simulations to predict the membrane permeability of natural drugs—withanolides (

Ashwagandha Root Extract and Its Potential Modulation of CCN1‐Associated Pathways in Sensitive‐Skin Aging [PDF]

open access: yesJ Cosmet Dermatol
ABSTRACT Background Cellular Communication Network Factor 1 (CCN1) has been identified as a key regulator of oxidative stress‐induced skin aging and sensitivity. Ashwagandha root extract (ARE) is a promising candidate for modulating CCN1‐associated pathways. Aims To investigate whether ARE can mitigate sensitive‐skin aging by modulating CCN1 expression

Deciphering Antidiabetic, Anti‐Inflammatory, and Anti‐Cholinergic Bioactivities of Withania somnifera (L) Dunal Root Extract: An In Vitro and In Silico Investigation

open access: yesChemistry &Biodiversity, Volume 23, Issue 6, June 2026.
In this investigation, Withania somnifera root ethanol extract displays anti‐inflammatory and antidiabetic properties. Though compounds including withasomniferolide B and sitoindoside IX have been previously identified as phytoconstituents in the plant, in silico analysis from this study revealed them as new lead compounds that could act as potential ...

Wild type p53 function in p53Y220C mutant harboring cells by treatment with Ashwagandha derived anticancer withanolides: bioinformatics and experimental evidence

open access: yesJournal of Experimental & Clinical Cancer Research, 2019
Background Tumor suppressor p53 protein is frequently mutated in a large majority of cancers. These mutations induce local or global changes in protein structure thereby affecting its binding to DNA.
