25-Hydroxycholesterol acts as an amplifier of inflammatory signaling [PDF]

open access: yesProceedings of the National Academy of Sciences, 2014
Significance The lipid 25-hydroxycholesterol (25HC) is produced by immune cells in response to infection. We demonstrate that in addition to interfering with viral entry and replication, 25HC also amplifies the activation of immune cells and increases the production of immune mediators.

The convenient synthesis of 25-hydroxycholesterol.

open access: yesChemical and Pharmaceutical Bulletin, 1979
The convenient synthesis of 25-hydroxycholesterol (6) is described. 5β-Cholestan-3α, 25-diol (2) derived from lithocholic acid was converted to 6 via four steps : oxidationbromination, dehydrobromination, deconjugation, and reduction. The overall yield of 25-hydroxycholesterol (6) starting from 2 was ca. 50%.

24S-hydroxycholesterol and 25-hydroxycholesterol differentially impact hippocampal neuronal survival following oxygen-glucose deprivation. [PDF]

open access: yesPLoS ONE, 2017
N-methyl-D-aspartate receptors (NMDARs), a major subtype of glutamate receptor mediating excitatory transmission throughout the CNS, participate in ischemia-induced neuronal death.

Marked upregulation of cholesterol 25-hydroxylase expression by lipopolysaccharide

open access: yesJournal of Lipid Research, 2009
During screening of genes upregulated by lipopolysaccharide (LPS; endotoxin) treatment of bone marrow-derived mouse macrophages, it was unexpectedly found that cholesterol 25-hydroxylase (Ch25h) was strongly upregulated.

Biosynthesis of the regulatory oxysterol, 5-cholesten-3β,25-diol 3-sulfate, in hepatocytes

open access: yesJournal of Lipid Research, 2007
Cellular cholesterol homeostasis is maintained through coordinated regulation of cholesterol synthesis, degradation, and secretion. Nuclear receptors for oxygenated cholesterol derivatives (oxysterols) are known to play key roles in the regulation of ...

Regulation of endoplasmic reticulum cholesterol by plasma membrane cholesterol

open access: yesJournal of Lipid Research, 1999
The abundance of cell cholesterol is governed by multiple regulatory proteins in the endoplasmic reticulum (ER) which, in turn, are under the control of the cholesterol in that organelle.

Highly sensitive quantification of key regulatory oxysterols in biological samples by LC-ESI-MS/MSs⃞

open access: yesJournal of Lipid Research, 2009
We describe a highly sensitive and specific method for the quantification of key regulatory oxysterols in biological samples. This method is based upon a stable isotope dilution technique by liquid chromatography-tandem mass spectrometry (LC-MS/MS ...

Metabolism of 25-hydroxycholesterol in mammalian cell cultures. Side-chain scission to pregnenolone in mouse L929 fibroblasts.

open access: yesJournal of Lipid Research, 1989
Three mammalian cell lines were examined for their ability to metabolize the regulatory oxysterol, 25-hydroxycholesterol, and derepress 3-hydroxy-3-methylglutaryl CoA reductase.

Spermine synthesis inhibitor blocks 25-hydroxycholesterol-induced- apoptosis via SREBP2 upregulation in DLD-1 cell spheroids

open access: yesBiochemistry and Biophysics Reports, 2020
The oxysterol 25-hydroxycholesterol (25-HC) has diverse physiological activities, including the ability to inhibit anchorage-independent growth of colorectal cancer cells. Here, we found that a polyamine synthesis inhibitor, DFMO, prevented 25-HC-induced

Neuronal Cholesterol Deficiency Mediated by Astrocytic SREBP2 Downregulation Leads to Postoperative Cognitive Dysfunction Through Impairment of Hippocampal Synaptic Plasticity and Excitatory Synaptic Transmission

open access: yesAdvanced Science, EarlyView.
The results showed that the reduction of plasma 24‐hydroxycholesterol (24‐OHC) was related to the cognitive decline in elderly surgical patients. Neuroinflammation induced by anesthesia and surgery significantly downregulated SREBP2 in hippocampal reactive astrocytes, resulting in reduced cholesterol synthesis.
