The Cross-Talk Between the Peripheral and Brain Cholesterol Metabolisms

open access: yesCurrent Issues in Molecular Biology
Cholesterol is an essential element for the development and normal function of the central nervous system. While peripheral cholesterol is influenced by liver metabolism and diet, brain cholesterol metabolism takes place in an isolated system due to the ...

On the substrate specificity of human CYP27A1

open access: yesJournal of Lipid Research, 2003
The mitochondrial sterol 27-hydroxylase (CYP27A1) is required for degradation of the C27-sterol side chain in bile acid biosynthesis. CYP27A1 seems, however, to have roles beyond this, as illustrated by patients with a deficient sterol 27-hydroxylase due

A phase 3, randomized clinical trial of soticlestat as adjunctive therapy for Lennox–Gastaut syndrome

open access: yesEpilepsia, EarlyView.
Soticlestat as adjunctive therapy for Lennox–Gastaut syndrome. Abstract Objective There remains a need for new treatments for Lennox–Gastaut syndrome (LGS), a developmental and epileptic encephalopathy with a heterogenous patient population that often requires polytherapy. The phase 3, randomized SKYWAY study (NCT04938427) investigated the efficacy and

Differential effects of 24-hydroxycholesterol and 27-hydroxycholesterol on β-amyloid precursor protein levels and processing in human neuroblastoma SH-SY5Y cells [PDF]

open access: yes, 2009
Background Activation of the liver × receptors (LXRs) by exogenous ligands stimulates the degradation of β-amyloid 1–42 (Aβ42), a peptide that plays a central role in the pathogenesis of Alzheimer's disease (AD).

Changes in the gut microbiome during fasting‐induced molting and its effects on gut–liver function

open access: yesiMetaOmics, EarlyView.
During fasting, harmful microbiota and metabolites rise, triggering intestinal injury and activation of the TGF‐β signaling pathway, which enhances intestinal stem cell proliferation. Concurrently, hepatic function declines, indicated by increased hepatic and serum bile acids and upregulation of the non‐classical bile acid synthesis pathway.

Increased appearance rate of 27-hydroxycholesterol in vivo in hypercholesterolemia: A possible compensatory mechanism.

open access: yes, 2012
BACKGROUND AND AIMS: The first step in the alternative pathway of bile acid biosynthesis is the 27-hydroxylation of cholesterol, which takes place both in liver and extrahepatic tissues.

Bile acid synthesis: 7 alpha-hydroxylation of intermediates in the sterol 27-hydroxylase metabolic pathway.

open access: yesJournal of Lipid Research, 1996
The recognition that the 7 alpha-hydroxylation of 27-hydroxycholesterol is catalyzed by an enzyme that is different from cholesterol 7 alpha-hydroxylase raises the question as to the number of similar enzymes that may be present in liver and subserve ...

Biomimetic mineralization‐inspired formulation‐adjuvant integrated circRNA nanovaccine against COVID‐19

open access: yesInterdisciplinary Medicine, EarlyView.
Inspired by the role of Mg2+ in stabilizing RNA secondary structures, we developed a one‐pot biomimetic mineralization method for synthesizing a circular RNA‐based nanovaccine (circRNA@FAiNVac) that integrates formulation and adjuvant functions. This nanovaccine exhibits potent immunostimulatory activation enabling multimodal immune responses and ...

Metabolism of 4 beta -hydroxycholesterol in humans

open access: yes, 2002
One of the major oxysterols in the human circulation is 4 beta-hydroxycholesterol formed from cholesterol by the drug-metabolizing enzyme cytochrome P450 3A4.

Simultaneous determination of oxysterols, cholesterol and 25-hydroxy-vitamin D3 in human plasma by LC-UV-MS. [PDF]

open access: yesPLoS ONE, 2015
Oxysterols are promising biomarkers of neurodegenerative diseases that are linked with cholesterol and vitamin D metabolism. There is an unmet need for methods capable of sensitive, and simultaneous quantitation of multiple oxysterols, vitamin D and ...
