Caloric restriction reveals a metabolomic and lipidomic signature in liver of male mice [PDF]

open access: yes, 2014
Lipid composition, particularly membrane unsaturation, has been proposed as being a lifespan determinant, but it is currently unknown whether caloric restriction (CR), an accepted life-extending intervention, affects cellular lipid profiles.

Polypeptide Modification and Cross-Linking by Oxidized 3-Hydroxykynurenine [PDF]

open access: yesBiochemistry, 2000
3-Hydroxykynurenine (3OHKyn) is present in the mammalian lens as a UV filter and is formed from kynurenine in the tryptophan metabolic pathway. 3OHKyn is a readily autoxidized o-aminophenol which binds to proteins in vitro. The lens, particularly its central region, the nucleus, becomes increasingly oxidized with age.

Increased indoleamine-2,3-dioxygenase activity is associated with poor clinical outcome in adults hospitalized with influenza in the INSIGHT FLU003Plus study [PDF]

open access: yes, 2017
BACKGROUND: Indoleamine-2,3-dioxygenase (IDO) mediated tryptophan (TRP) depletion has antimicrobial and immuno-regulatory effects. Increased kynurenine (KYN)-to-TRP (KT) ratios, reflecting increased IDO activity, have been associated with poorer ...

Gourami Lenses Convert Tryptophan into 3-Hydroxykynurenine

open access: yesOphthalmic Research, 2009
Although free tryptophan was found in the lens of both the kissing gourami (Helostoma temmincki) and the 3-spot gourami (Trichogaster trichopterus), only the lens of the latter species contained 3-hydroxykynurenine. Radiolabel experiments carried out on 3-spot gourami lenses in culture demonstrated that, as in humans, lenticular tryptophan is converted

Indoleamine-2,3-dioxygenase activity in experimental human endotoxemia [PDF]

open access: yes, 2013
Background: Excessive tryptophan metabolism to kynurenine by the rate-limiting enzyme endothelial indoleamine 2,3-dioxygenase 1 (IDO) controls arterial vessel relaxation and causes hypotension in murine endotoxemia.

Kynurenines link chronic inflammation to functional decline and physical frailty

open access: yesJCI Insight, 2020
Chronic inflammation is associated with physical frailty and functional decline in older adults; however, the molecular mechanisms of this linkage are not understood.

Fast and low-cost evaluation of hydroxykynurenine activity

open access: yesMethodsX, 2020
The enzyme 3-hydroxykynurenine transaminase (HKT) acts as an important enzyme in tryptophan catabolism of disease-carrier insects, e.g. Aedes aegypti and Anopheles gambiae. HKT is a detoxification enzyme that converts 3-hydroxykynurenine (a precursor for
