Proteinase-activated receptors: structural requirements for activity, receptor cross-reactivity, and receptor selectivity of receptor-activating peptides

Canadian Journal of Physiology and Pharmacology, 1997
We have used three distinct bioassay systems (rat aorta (RA) relaxation; rat gastric longitudinal muscle (LM) contraction; human embryonic kidney 293 (HEK293) cell calcium signal) to evaluate the activity and receptor selectivity of analogues of the receptor-activating peptides derived either from the thrombin receptor (TRAPs, based on the human ...

The activation of the CGRP receptor

Biochemical Society Transactions, 2013
The CGRP (calcitonin gene-related peptide) receptor is a family B GPCR (G-protein-coupled receptor). It consists of a GPCR, CLR (calcitonin receptor-like receptor) and an accessory protein, RAMP1 (receptor activity modifying protein 1). RAMP1 is needed for CGRP binding and also cell-surface expression of CLR.

Proteinase-Activated Receptors

Pharmacological Reviews, 2001
Proteinase-activated receptors are a recently described, novel family of seven-transmembrane G-protein-coupled receptors. Rather then being stimulated through ligand receptor occupancy, activation is initiated by cleavage of the N terminus of the receptor by a serine protease resulting in the generation of a new tethered ligand that interacts with the ...

Activation of membrane receptors

Endocrine, 1995
Many extracellular messengers interact with discriminate receptors on the cell surface. Some of bound ligands activate receptors whereas others fail to do so. Only activated receptors are capable of generating and transferring signal through the membrane.

Platelet-activating factor receptor

Prostaglandins & Other Lipid Mediators, 2002
Platelet-activating factor (PAF, 1-O-alkyl-2-acetyl-sn-glycero-3-phosphocholine) is a biologically active phospholipid mediator. Although PAF was named for its potential to induce platelet aggregation, intense investigations have elucidated potent biological actions of PAF in a broad range of cell types and tissues.

The receptor for urokinase‐plasminogen activator

Journal of Cellular Biochemistry, 1986
AbstractMany human cells and cell lines possess a specific receptor that binds urokinase plasminogen activator (uPA) with an affinity of about 10−10 M. Bound enzyme is not internalized, is slowly dissociated, and retains its enzymatic activity. The amino acid sequence of uPA responsible for receptor binding is located within the first 35 aminoterminal ...

Regulation of glucocorticoid receptor activity

Journal of Steroid Biochemistry, 1989
Four levels of regulation of glucocorticoid receptor (GR) activity have been investigated. (1) Phosphorylation of the GR was studied in NIH 3T3 cells metabolically labeled with [32P]orthophosphate. A highly specific antiserum against the GR was used to immunoprecipitate 32P-labeled GR, and protein blotting was used to determine the GR concentration ...

Constitutive Activity in Gonadotropin Receptors

2014
Constitutively active mutants (CAMs) of gonadotropin receptors are, in general, rare conditions. Luteinizing hormone-choriogonadotropin receptor (LHCGR) CAMs provoke the dramatic phenotype of familial gonadotropin-independent isosexual male-limited precocious puberty, whereas in females, there is not yet any identified phenotype.

Trk neurotrophin receptor activators

Drug News & Perspectives, 2002
The neurotrophins are a small group of proteins whose binding at the tropomyosin related kinase (Trk) and p75 neurotrophin receptor transmembrane receptors triggers a cascade of signaling events that give rise to neurotrophic responses in neuronal cells and in vivo.
