The mammalian target of rapamycin complex 1 (mTORC1) is a master regulator of cell growth that is commonly deregulated in human diseases. Here we find that mTORC1 controls a transcriptional program encoding amino acid transporters and metabolic enzymes ...

A GSH‐responsive AIE nanoplatform (TSH NPs) with RGD‐mediated tumor targeting co‐delivers a photosensitizer (TPA) and HCPT for extrahepatic cholangiocarcinoma. The system synergistically enhances PDT and chemotherapy by suppressing HIF‐1α/VEGF‐driven hypoxic adaptation, effectively overcoming PDT resistance.

Hepatic metabolism is critical for the maintenance of whole body glucose and lipid metabolism. Excessive hepatic glucose production and de novo lipogenesis contributes to fasting hyperglycemia in diabetes and hepatic steatosis, respectively. Common genetic variants of the fat mass and obesity associated (FTO) gene are strongly associated with obesity ...

This study uncovers that quercetin naturally targets mitochondria. By coordinating quercetin with Fe3+, we engineer an ultrasmall cascade nanozyme (MCN) with superoxide dismutase‐catalase activities. MCN crosses the damaged blood–brain barrier, scavenges mitochondrial ROS, prevents mitochondrial DNA leakage, and blocks the cGAS‐STING pathway, thereby ...

Spatiotemporal Targeting Randle Cycle and Immune Checkpoint for Potent Antitumor Therapy
A glucose oxidase‐based nanogel (GOX‐NG) system using catechol‐functionalized alginate, exhibits enhanced tumor penetration, prolonged retention, and sustained glucose depletion in the tumor microenvironment. When combined with a fatty acid oxidation inhibitor, it implements dual metabolic suppression, thereby enhancing ROS‐induced immunogenic cell ...

Inhibition of KDELR2 in a small fraction of tumor cells generates sustainable immunogenic cell death conditions within the tumor microenvironment. These conditions promote the regression of tumors in a T cell independent manner. During regression, macrophages prime T cells that subsequently provide systemic protection against recurrence. The potency of
