Molecular Mechanisms and Probe‐Dependent Effects of Clinically Relevant GABAA Receptor Modulators
Combining molecular dynamics, dynamic pharmacophores, and allosteric network analysis, we reveal that benzodiazepines and Z‐drugs stabilize GABA binding through long‐range TMD–ECD allosteric coupling, while anesthetics directly activate the channel via unique L232/I264 contacts, bypassing GABA.
