Tolerance and antagonism to allopregnanolone effects in the rat CNS [PDF]

open access: yes, 2006
Many studies have suggested a relationship between sex steroids and negative mental and mood changes in women. Allopregnanolone, a potent endogenous ligand of the GABA-A receptor and a metabolite of progesterone, is one of the most accused neuroactive ...

Pediatric super‐refractory status epilepticus treated with allopregnanolone [PDF]

open access: yes, 2014
Super-refractory status epilepticus is a life-threatening condition. Resistance to benzodiazepine and barbiturate treatment for this disorder is thought to be due to internalization of synaptic γ-aminobutyric acid (GABA)A receptors, and withdrawal of ...

Divergent Transcriptomic Effects of Allopregnanolone in Postpartum Depression

open access: yesGenes, 2023
Brexanolone, a formulation of the neurosteroid allopregnanolone (ALLO), is approved for treating postpartum depression (PPD) and is being investigated for therapeutic efficacy across numerous neuropsychiatric disorders. Given ALLO’s beneficial effects on mood in women with PPD compared to healthy control women, we sought to characterize and compare the

The estrous cycle modulates early-life adversity effects on mouse avoidance behavior through progesterone signaling

open access: yesNature Communications, 2022
Early-life adversity (ELA) increases the likelihood of neuropsychiatric diagnoses, which are more prevalent in women than men. Since changes in reproductive hormone levels can also increase the probability of anxiety disorders in women, we examined the ...

Molecular Mechanisms and Probe‐Dependent Effects of Clinically Relevant GABAA Receptor Modulators

open access: yesArchiv der Pharmazie, Volume 359, Issue 6, June 2026.
Combining molecular dynamics, dynamic pharmacophores, and allosteric network analysis, we reveal that benzodiazepines and Z‐drugs stabilize GABA binding through long‐range TMD–ECD allosteric coupling, while anesthetics directly activate the channel via unique L232/I264 contacts, bypassing GABA.

Alfaxalone anaesthesia increases brain derived neurotrophic factor levels and preserves postoperative cognition by activating pregnane-X receptors: an in vitro study and a double blind randomised controlled trial

open access: yesBMC Anesthesiology, 2022
Background Alfaxalone is a fast acting intravenous anaesthetic with high therapeutic index. It is an analogue of the naturally-occurring neurosteroid allopregnanolone responsible for maintenance of cognition and neuroprotection by activation of brain ...

Women with epilepsy: Evidence‐based counseling across the lifespan

open access: yesEpilepsia, Volume 67, Issue 6, Page 2689-2706, June 2026.
Abstract Women with epilepsy (WWE) encounter distinct and evolving challenges across the lifespan that require clinical management extending beyond seizure control alone. Although awareness of sex‐specific aspects of epilepsy has increased, important gaps remain in their integration into routine care.
