Engineered extracellular vesicles enriched with the miR‐214/199a cluster enhance the efficacy of chemotherapy in ovarian cancer

open access: yesMolecular Oncology, EarlyView.
Loss of the miR‐214/199a cluster is associated with recurrence in ovarian cancer. Engineered small extracellular vesicles (m214‐sEVs) elevate miR‐214‐3p/miR‐199a‐5p in tumor cells, suppress β‐catenin, TLR4, and YKT6 signaling, reprogram tumor‐derived sEV cargo, reduce chemoresistance and migration, and enhance carboplatin efficacy and survival in ...

Recent Developments in Alternatives to Animal Testing [PDF]

open access: yes, 2019
Methods that replace techniques that use live animals, or methods of testing substances without live animal use, are known as alternatives, replacements or non-animal methods. Some prefer the term advanced technologies given the fact that they often rely

Navigating the unseen peril: safeguarding medical imaging in the age of AI

open access: yesFrontiers in Artificial Intelligence
In response to the increasing significance of artificial intelligence (AI) in healthcare, there has been increased attention – including a Presidential executive order to create an AI Safety Institute – to the potential threats posed by AI.

Interrogating the immune landscape of microsatellite stable RAS‐mutated colon cancer

open access: yesMolecular Oncology, EarlyView.
COLOSSUS project RAS‐mutated MSS colon cancer study explored transcriptomics and immune cell density by immunohistochemistry (IHC), Immunoscore (IS), ISIC/TuLIS scores, mutation counts, and detected different prevalences but similar microenvironment composition across immune markers with clinical relevance for future immunotherapy combination ...

Towards an in vitro test system for developmental neurotoxicity: a new test method using mouse embryonic stem cells [PDF]

open access: yes, 2007
Today, on a large number of chemicals we lack information on their adverse effects on the developing nervous system. Existing in vivo test methods are time consuming, laborious, expensive and require high numbers of laboratory animals. Thus, standardized,

Prediction of Chemical Respiratory and Contact Sensitizers by OX40L Expression in Dendritic Cells Using a Novel 3D Coculture System

open access: yesFrontiers in Immunology, 2017
The use of animal models in chemical safety testing will be significantly limited due to the recent introduction of the 3Rs principle of animal experimentation in research.

Keratin 19 as a prognostic marker and contributing factor of metastasis and chemoresistance in high‐grade serous ovarian cancer

open access: yesMolecular Oncology, EarlyView.
Keratin 19 (KRT19) is overexpressed in high‐grade serous ovarian cancer with high levels of Kallikrein‐related peptidases (KLK) 4–7 and is associated with poor survival. In vivo analyses demonstrate that elevated KRT19 increases peritoneal tumour burden.

New trends on alternative to animal testings in Japan

open access: yesFolia Pharmacologica Japonica, 2018
Non-animal alternative test methods have developed in the field of genotoxicity, endocrine disrupter, irritation and skin sensitization, and these are used the safety evaluation of chemical, pesticide, pharmaceutical and cosmetic for regulatory use. For the systemic toxicological endpoints of repeated dose toxicity, carcinogenicity, immunotoxicity and ...

Establishment of a humanized patient‐derived xenograft mouse model of high‐grade serous ovarian cancer for preclinical evaluation of combination immunotherapy

open access: yesMolecular Oncology, EarlyView.
We have established a humanized orthotopic patient‐derived xenograft (Hu‐oPDX) mouse model of high‐grade serous ovarian cancer (HGSOC) that recapitulates human tumor–immune interactions. Using combined anti‐PD‐L1/anti‐CD73 immunotherapy, we demonstrate the model's improved biological relevance and enhanced translational value for preclinical ...
