Anti-nociceptive and Anti-inflammatory Activities of Wrightia arborea

open access: yesPakistan Journal of Biological Sciences, 2013
Anti-nociceptive and anti-inflammatory effects of methanolic extract of Wrightia arborea (MEWA) were examined using different models in rats. MEWA was given to rats orally upto 2000 mg kg(-1) b.wt. for acute toxicity study and observed for 14 days. Anti-nociceptive activity was evaluated in rats against Acetic acid induced writhing (chemically induced ...

Decoding the Neural Circuit Underlying Context‐Cue Association in Retrieval of Morphine Reward Memory

open access: yesAdvanced Science, EarlyView.
Contextual cues recruit the glutamatergic vCA1–dmPFC pathway to retrieve morphine reward memory by reshaping cell‐type‐specific prefrontal microcircuit activity. Distinct regulation of PV+ and SST+ interneurons defines this hippocampal–prefrontal mechanism.

Astrocytes Coordinate Neurovascular Coupling During Prolonged Sensory Demand via AQP4/TRPA1/BEST1/D‐Serine Signaling

open access: yesAdvanced Science, EarlyView.
Neuronal activation leads to astrocytic transient volume changes, activating transient receptor potential ankyrin 1 (TRPA1) channels, leading to Ca2+ influx. Activation of TRPA1 causes the release of D‐serine through BEST1. Finally, the augmented extracellular levels of D‐serine boost neuronal activity by binding GluN1‐containing NMDARs, leading to CBV

Synthesis and anti-nociceptive potential of isoxazole carboxamide derivatives

open access: yesBMC Chemistry, 2019
Isoxazole is an important pharmacophore in medicinal chemistry with a wide range of pharmacological activities. The present study deals with the synthesis and evaluation of antinociceptive potential of nine novel 3-substituted-isoxazole-4-carboxamide derivatives. In the first step, respective oxime was prepared and further treated with ethylacetoacetate

STING/Type I Interferon Pathway Activation Confers Multimodal Therapeutic Benefits in Osteomyelitis

open access: yesAdvanced Science, EarlyView.
STING/type I interferon signaling counteracts osteomyelitis through coordinated control of pain, bone destruction, and abscess formation. It inhibits pathogen‐induced nociceptor activation, restrains osteoclast‐mediated bone resorption, and suppresses IL‐1β‐driven neutrophil recruitment.

Cannabinoid CB1 Receptor in Nociceptors Mediates Postoperative Analgesia via ASIC3 Inhibition

open access: yesAdvanced Science, EarlyView.
CB1R and ASIC3 are co‐expressed in CGRP+ nociceptors of mouse DRG. Plantar incision triggers an upregulation of ASIC3 and activation of ASIC3+ neurons, driving mechanical allodynia. CB1R activation suppresses ASIC3 function and downstream ERK1/2 phosphorylation, thereby braking nociceptive signaling and accelerating pain resolution.
