Scaffold Hopping Provides Liver X Receptor Agonist With Preference for the β Subtype
A scaffold hopping strategy was pursued to enhance the binding pocket engagement and β‐subtype selectivity of liver X receptor agonist 2. The resulting spirocyclic lead compound, (S)‐7, demonstrates superior potency, enhanced potency‐based selectivity for LXRβ, and improved microsomal stability over the existing scaffold.

High Density Lipoproteins Inhibit Oxidative Stress-Induced Prostate Cancer Cell Proliferation
Recent evidence suggests that oxidative stress can play a role in the pathogenesis and the progression of prostate cancer (PCa). Reactive oxygen species (ROS) generation is higher in PCa cells compared to normal prostate epithelial cells and this ...

Metabolite‐driven redox stress governs cancer cell senescence. TPI1 deficiency elevates DHAP, initiating SARM1‐dependent cADPR‐Ca2+ release. Mitochondrial ROS surge subsequently induces DNA damage and senescence, offering a conserved therapeutic target in multiple cancers.

Genetic ablation of Cep55 in Pten‐deficient mouse models delays tumorigenesis. Integrated multi‐omics analyses (proteomics, phosphoproteomics, and spatial transcriptomics) reveal that CEP55 regulates oncogenic signaling (RAS/ERK, PI3K/AKT), integrin/FAK‐mediated adhesion, extracellular matrix (ECM) remodeling, and endocytosis.

ABCB5 and the regulation of p16INK4a by non-coding RNA [PDF]
p16INK4a (p16) traps the cell at the restriction point of the cell cycle by binding to cyclin-dependent kinase 4/6 thus preventing the phosphorylation of the retinoblastoma protein (pRB).

Associations of plasma HDL subclasses with incident dementia risk in older adults
Abstract INTRODUCTION Plasma high‐density lipoprotein (HDL) varies in size. We examined the associations between HDL subclasses and their lipid composition and incident dementia risk in older adults. METHODS We included 23,790 United Kingdom (UK) Biobank (UKB) and 147 Atherosclerosis Risk in Communities (ARIC) older adults free of dementia at baseline.
