Light-Activating PROTACs in Cancer: Chemical Design, Challenges, and Applications

open access: yesApplied Sciences, 2022
Nonselective cell damage remains a significant limitation of radiation therapies in cancer. Decades of successful integration of radiation therapies with other medicinal chemistry strategies significantly improved therapeutic benefits in cancer ...

TP53 R249S mutation in hepatic organoids captures the predisposing cancer risk

open access: yesHepatology, EarlyView., 2022
The systematic approach in elucidating the gain‐of‐function (GOF) roles of TP53 mutations in early liver carcinogenesis. Unique downstream targets of TP53 L3 mutations were identified from chormatin immunoprecipitation sequencing in HCC cell lines, followed by a series of validation assays to substantiate the exclusive transcriptional regulations ...

Rational design, synthesis and in vitro evaluation of cell-active chemical probes for bromodomain-containing proteins Alessandra [PDF]

open access: yes, 2021
2018 - 2019Bromodomains (BRDs) are epigenetic readers able to selectively recognize the acetylated lysine residues on histone and non-histone proteins.

Duality in bromodomain-containing protein complexes [PDF]

open access: yesFrontiers in Bioscience, 2001
Proteins that contain a motif called a bromodomain are implicated in both transcriptional activation and repression. The bromodomain of p/CAF, the only solution structure of a bromodomain that has been solved to date, reveals that the motif binds N-acetyl-lysine groups, presumably to anchor enzymatic functions to histones and by extension to chromatin.

Bromodomain-Containing Protein 2 gene in photosensitive epilepsy [PDF]

open access: yesSeizure, 2012
Photosensitive epilepsy (PSE) is a form of reflex epilepsy characterized by seizures triggered by light. Genetic factors play an important role and some studies have indicated a possible role of the bromodomain-containing protein 2 (BRD2) gene. Our aim was to investigate the relationship between PSE and mutations of the BRD2 gene.Fifty-four PSE ...

Genome-wide analysis of bromodomain gene family in Arabidopsis and rice

open access: yesFrontiers in Plant Science, 2023
The bromodomain-containing proteins (BRD-proteins) belongs to family of ‘epigenetic mark readers’, integral to epigenetic regulation. The BRD-members contain a conserved ‘bromodomain’ (BRD/BRD-fold: interacts with acetylated-lysine in histones), and ...

Dissecting the Role of BET Bromodomain Proteins BRD2 and BRD4 in Human NK Cell Function

open access: yesFrontiers in Immunology, 2021
Natural killer (NK) cells are innate lymphocytes that play a pivotal role in the immune surveillance and elimination of transformed or virally infected cells.

BRD3 (bromodomain containing 3) [PDF]

open access: yes, 2016
BRD3 is a ubiquitously expressed member of the bromodomain and extraterminal motif (BET) family of proteins that use their tandem N-terminal bromodomains to associate with acetylated histones and transcription factors.

Androgen Receptor Deregulation Drives Bromodomain-Mediated Chromatin Alterations in Prostate Cancer

open access: yesCell Reports, 2017
Global changes in chromatin accessibility may drive cancer progression by reprogramming transcription factor (TF) binding. In addition, histone acetylation readers such as bromodomain-containing protein 4 (BRD4) have been shown to associate with these ...

Integrated genomic analysis of the 8q24 amplification in endometrial cancers identifies ATAD2 as essential to MYC-dependent cancers. [PDF]

open access: yesPLoS ONE, 2013
Chromosome 8q24 is the most commonly amplified region across multiple cancer types, and the typical length of the amplification suggests that it may target additional genes to MYC.
