Delivery of Pleckstrin‐Homology Domains Suppresses PI3K/Akt Signaling and Breast Cancer Metastasis

open access: yesAdvanced Science, EarlyView.
Current therapies curb tumor growth but not metastasis. Obscurin, a giant metastasis suppressor lost in breast cancer, restrains PI3K/Akt signaling but is impractical to restore. We deploy a mini‐obscurin, comprising the obscurin‐PH‐domain, which sequesters PI3K‐p85, potently suppressing invasion and metastasis.

Dexamethasone in glioblastoma multiforme therapy : mechanisms and controversies [PDF]

open access: yes, 2019
Glioblastoma multiforme (GBM) is the most common and malignant of the glial tumors. The world-wide estimates of new cases and deaths annually are remarkable, making GBM a crucial public health issue.

Proton FLASH Exposure Preserves Gut Commensal Microbiomes and Spares Intestinal Stem Cells

open access: yesAdvanced Science, EarlyView.
This study highlights the role of Proton FLASH abdominal irradiation in sparing of intestinal stem cells and preservation of key gut microbial population resulting minimization of radiation toxicity in intestinal epithelium in mice. Our findings support the potential of Proton FLASH to improve the therapeutic ratio for abdominal radiation exposure ...

Structural Basis of Clostridium perfringens Enterotoxin Activation and Oligomerization by Trypsin

open access: yesToxins, 2023
Clostridium perfringens enterotoxin (CpE) is a β-pore forming toxin that disrupts gastrointestinal homeostasis in mammals by binding membrane protein receptors called claudins.

Claudin-containing exosomes in the peripheral circulation of women with ovarian cancer [PDF]

open access: yes, 2009
Background The absence of highly sensitive and specific serum biomarkers makes mass screening for ovarian cancer impossible. The claudin proteins are frequently overexpressed in ovarian cancers, but their potential as prognostic, diagnostic, or detection

Methylglyoxal Accumulation is Associated with Brain Inflammation after Myocardial Infarction with Sex and Regional Differences

open access: yesAdvanced Science, EarlyView.
This study identifies that methylglyoxal may play an important role in heart‐brain interactions after myocardial infarction. Myocardial infarction leads to increased levels of methylglyoxal‐derived advanced glycation end‐products (MG‐H1) in the brain of mice, which is associated with loss of blood‐brain barrier integrity and neuroinflammation ...

Salinity Regulates Claudin mRNA and Protein Expression in the Teleost Gill [PDF]

open access: yes, 2008
Udgivelsesdato: 2008-Jan-9The teleost gill carries out NaCl uptake in fresh water (FW) and NaCl excretion in seawater (SW). This transformation with salinity requires close regulation of ion transporter capacity and epithelial permeability.

A Synergistic Hydrogel‐Microalgae Platform for Dual‐Targeting of Intestinal and Neuroimmune Dysfunction in Inflammatory Bowel Disease

open access: yesAdvanced Science, EarlyView.
A bioactive hydrogel incorporating Chlorella vulgaris and paeoniflorin within a carboxymethyl chitosan/sodium alginate matrix cross‐linked with genipin is developed for inflammatory bowel disease therapy. The hydrogel exhibits gastric stability and intestine‐responsive release.

Structural and biophysical insights into targeting of claudin-4 by a synthetic antibody fragment

open access: yesCommunications Biology
Claudins are a 27-member family of ~25 kDa membrane proteins that integrate into tight junctions to form molecular barriers at the paracellular spaces between endothelial and epithelial cells.

Mouse Models of Human Claudin-Associated Disorders: Benefits and Limitations [PDF]

open access: yes, 2019
In higher organisms, epithelia separate compartments in order to guarantee their proper function. Such structures are able to seal but also to allow substances to pass.
