Structural studies and functional engineering of NanX: an anhydro‐sialic acid transporter from Escherichia coli

open access: yesFEBS Open Bio, EarlyView.
Biophysical characterisation shows that NanX, a membrane transport protein from the major facilitator superfamily (MFS), forms both monomers and dimers after purification. AlphaFold modelling and substrate docking provide information on residues likely involved in substrate recognition for NanX and another MFS member, NanT.

Cryo-Electron Tomography in Plant Biology

open access: yesAnnual Review of Plant Biology
Cryo-electron tomography (cryo-ET) is a transformative technique in cell biology that enables three-dimensional visualization of cellular structures in near-native states and at nanometer and even subnanometer resolution. Unlike traditional imaging methods, cryo-ET preserves the ultrastructure of cells without chemical fixation or staining, allowing ...

Parallel cryo electron tomography on in situ lamellae

open access: yesNature Methods, 2022
In situ cryo electron tomography of cryo focused ion beam milled samples emerged in recent years as a powerful technique for structural studies of macromolecular complexes in their native cellular environment. The lamella-shaped samples, however, have a limited area and are created with a necessary pretilt.

Comparative assessment of crystallographic and cryo‐EM models in the Protein Data Bank

open access: yesFEBS Open Bio, EarlyView.
Raw data obtained by X‐ray crystallography or cryo‐EM result in experimental maps, ultimately fitted by atomic models. Although the physical principles are different, the final results can be viewed, compared, and evaluated in the same way. With cryogenic electron microscopy (cryo‐EM) on track to surpass X‐ray crystallography as the preferred method ...

The C‐terminal domain of yeast Arginyltransferase1 is essential for its catalytic activity

open access: yesFEBS Open Bio, EarlyView.
Arginyltransferase 1 (Ate1), a eukaryotic enzyme, catalyses arginylation, transferring arginine from tRNA‐Arg to the amino terminus of the target protein. Overexpression of Ate1 in yeast is lethal and is dependent on arginylation. This study elucidates how mutations in the cofactor‐binding and active site of Ate1 and truncation of its structural ...

Fluorination: A strategy to tune the interactions of molecules and nanoscale systems with biological matter

open access: yesFEBS Open Bio, EarlyView.
This minireview explores fluorination as a key strategy to optimize therapeutic molecules and nanocarriers. By incorporating different fluorinated motifs—from single atoms to linear and branched chains—drug delivery, cellular uptake, and colloidal stability can be improved and fine‐tuned.

Cryo-ET data processing can be roughly divided into 3 blocks.

open access: yes, 2021
Preprocessing (top) transforms raw experimental data, usually in the form of multiframe micrographs, into 3D images called tomograms. Particle picking (middle) derives particle position and orientation information from tomograms as well as an initial ...

Structural Conservation and Adaptation of the Bacterial Flagella Motor

open access: yesBiomolecules, 2020
Many bacteria require flagella for the ability to move, survive, and cause infection. The flagellum is a complex nanomachine that has evolved to increase the fitness of each bacterium to diverse environments. Over several decades, molecular, biochemical,

Computational resources for cryo-electron tomography in Bsoft [PDF]

open access: yesJournal of Structural Biology, 2008
The Bsoft package [Heymann, J.B., Belnap, D.M., 2007. Bsoft: image processing and molecular modeling for electron microscopy. J. Struct. Biol. 157, 3-18] has been enhanced by adding utilities for processing electron tomographic (ET) data; in particular, cryo-ET data characterized by low contrast and high noise.

Spatially Regulated Lithium Plating via Li2O‐Rich Interphase Enables Durable Fast‐Charging Hybrid Lithium Batteries

open access: yesAdvanced Functional Materials, EarlyView.
A Li2O‐rich solid electrolyte interphase (SEI) is engineered on graphite through a propylene carbonate‐based electrolyte to spatially regulate Li plating. The homogeneous interphase lowers Li nucleation barriers, promotes conformal Li deposition around graphite particles, and suppresses dendritic accumulation, enabling durable hybrid Li‐ion/Li‐metal ...
