Correlative cryo-fluorescence light microscopy and cryo-electron tomography of streptomyces

open access: yes, 2014
Light microscopy and electron microscopy are complementary techniques that in a correlative approach enable identification and targeting of fluorescently labeled structures in situ for three-dimensional imaging at nanometer resolution.

Contrast transfer function correction applied to cryo-electron tomography and sub-tomogram averaging [PDF]

open access: yes, 2009
Cryo-electron tomography together with averaging of sub-tomograms containing identical particles can reveal the structure of proteins or protein complexes in their native environment.

Preferential Interfacial Engineering of Hydrogels via Interdigitated Nanoparticle Assembly

open access: yesAdvanced Functional Materials, EarlyView.
Robust surface functionalization of hydrogel surfaces is performed via interdigitated metal nanoparticle assembly, inducing strong affinity with the hydrogel matrix. The partially exposed nanoparticle domains act as interfacial nanobridges that mediate both chemical and physical bonding with diverse materials, including small molecules, fluorous ...

Beyond Thin Film Limits—Superhard and Hydrogen‐Free Amorphous Carbon Coatings With Over 100 µm Thickness

open access: yesAdvanced Functional Materials, EarlyView.
For the first time, an extreme‐thick laser‐arc PVD ta‐C (DLC) coating exceeding 100 µm is demonstrated, opening up new possibilities for ultra‐high load bearing capacity. This superhard amorphous carbon architecture combines >50% sp3 bonding, robust adhesion and self‐supporting performance, shielding substrates from plastic flow under extreme contact ...

Fast and accurate defocus modulation for improved tunability of cryo-EM experiments

open access: yesIUCrJ, 2020
Current data collection strategies in electron cryo-microscopy (cryo-EM) record multiframe movies with static optical settings. This limits the number of adjustable parameters that can be used to optimize the experiment.

Cryo-electron Tomography: An Overview

open access: yes, 2022
Viruses are macromolecular machineries which hijack cellular metabolism for replication. Viruses that are enveloped, comprise a large variety of DNA and RNA virus, many of which are notorious human or animal pathogen. Despite their importance, presence of lipid bilayers in their assembly had made most enveloped viruses too pleomorphic to be ...

Dynamic Observation of Lacy Pattern Formation in a Bone Tissue Model Using Liquid Phase Scanning Transmission Electron Microscopy

open access: yesAdvanced Functional Materials, EarlyView.
Here, we establish a low‐dose liquid‐phase electron microscopy workflow for real‐time observation of biological processes in hydrated conditions. Beam damage is reduced by imaging only 20% of the pixels and reconstructing the complete image using an inpainting algorithm.
