Novel (S)‐5′‐guanidinopropyl uridine analogs bearing 2′‐O‐methyl or 2′‐fluoro substitutions enhance nuclease resistance and mismatch discrimination while preserving A‐form RNA structure and potent RNAi activity. These modifications provide a promising strategy for improving the stability, specificity, and therapeutic potential of siRNA‐based medicines.

DNA architectures with aminonaphthalimides and cyanovinylenes as base surrogates form interstrand and intrastrand dimers showed that can be used as nucleic acid‐sensitive probes by their fluorescence turn‐on and red‐shift as readouts.
